EC-SOD在大肠杆菌中的可溶性表达 朱希强,袁勤生*(华东理工大学生物反应器工程国家重点实验室,上海 200237) 摘要:目的在大肠杆菌中表达可溶性EC-SOD。方法将表达质粒EC-pet28a(+)转入宿主菌BL21(DE3)plysS中进行表达,在低温下培养含表达质粒EC-pet28a(+)的宿主菌BL21(DE3),通过SDS电泳,观察其在超声上清中的表达。结果EC-SOD在BL21(DE3)plysS中表达,其量比在BL21(DE3)降低约20%,上清中有明显表达;在20℃和15℃培养时,EC-SOD在BL21(DE3)的超声上清中有表达。结论EC-SOD在BL21(DE3)plysS中或在BL21(DE3)中低温培养,均可实现部分上清中的表达。关键词:EC-SOD;BL21(DE3)plysS;基因表达

Abstract:ObjectiveTo express the resolution EC-SOD by E.coli.MethodsThe expression plasmid EC-pet28a(+) was transformed into BL21(DE3)plysS and BL21(DE3)containning expression plasmid EC-pet28a(+) was cultivated in low temperature. The expression of EC-SOD in supernatant was observed by SDS. ResultsThe expressed amount of EC-SOD by BL21(DE3)plysS was less about 30% than that by BL21(DE3), and it could express in supernatant obviously; EC-SOD could also express in supernatant by BL21(DE3)which was fermented in the temperature 20℃ and 15℃.ConclusionEC-SOD can express in supernatant by BL21(DE3)plysS or by BL21(DE3) fermented in low temperature.Key words: EC-SOD; BL21(DE3)plysS; gene expression
??????人胞外超氧化物歧化酶(EC-SOD)是目前已知的3种SOD同工酶之一,是含Cu,Zn金属离子的分泌型糖蛋白,主要在血管壁高效表达[1], 有显著降血糖,降血脂及降血压作用[2~4]。EC-SOD在组织中含量极少,分离纯化相当困难,因此进行基因克隆表达具有重要意义。我室贺华君等构建了EC-SOD的表达质粒,并在BL21(DE3)中进行了表达[5]。表达产物均为包涵体。由于EC-SOD是四亚基的金属酶,每个亚基中含有6个巯基,使包涵体复性率较低。为此我们尝试了将此基因在大肠杆菌中进行可溶性表达。??????首先,将EC-SOD在BL21(DE3)plysS中进行表达。BL21(DE3)plysS中含有质粒plysS,此质粒可低水平持续表达T7溶菌酶,降低了T7RNA聚合酶的量,从而使重组基因的表达水平下降。另将原工程菌在低温下培养,降低基因表达速度,使其有充分时间进行正确折叠。以上措施均使EC-SOD在菌体超声上清中获得了表达。
1材料、仪器与试剂1.1实验材料???EC-SOD基因工程菌由本课题组构建;质粒Pet-28a(+)和菌株BL21(DE3)由中科院上海生物化学研究所吴祥甫教授提供;菌株BL21(DE3)plysS由本校张慧展教授的实验室提供;种子培养基用LB培养基。
