高效液相色谱法分析黑米皮花青素及其 体内代谢产物 王 庆 ,‘郭红辉’,张名位2,夏效东’,邓 斌 ,‘夏 敏’,凌文华 ‘ (1.中山大学公共卫生学院,广东广州 510080; 2.广东省农业科学院农业部黑色食品研究重点开放实验室,广东广州 510640) 摘 要:建立分析黑米皮中花青素含量及人体内代谢产物的高效液相色谱方法。并研究了黑米皮中花青素种类、 含量及在人体内的代谢情况。样品经预处理后,以HypersilBDSC,:柱,4%磷酸水溶液、乙睛(V/V)等度洗脱 分离,520nm波长下检测、外标法定量。结果表明,黑米皮中花青素总含量约为2.31%,其中矢车菊素一3-葡萄 糖f}(Cy-3-g)占1.87%,芍药素一3-葡萄糖普(Pn-3-g)占0.44%.Cy-3-g在血浆中的最高浓度为21.4714.48ng/ml,出 现在口服黑米皮后1.5h,血浆中未检测到Pn-3-goCy-3-g在。-2h的尿液样品中的含量最高(390.78士69.28ng/ml), Pn-3-g在2-4h收集的尿液中含量最高(105.90士26.26ng/ml)o 关键词:花青素:黑米;代谢;高效液相色谱法 DeterminationAnthocyamnsrfomBlackRiceOutlayerandHumanMetabolitesbyHPLC WANGQing,GUOHong-hui,ZHANGMing-wei2,XIAXiao-dong,DENGBin, XIAMin,LNIGWen-hua (l.SchoolofPublicHealth,SunYat-senUniversity,Guangzhou 510080,China;2.KeyLabofBlackFood Research,MinistryofAgriculture,GuangdongAcademyofAgriculturalSciences,Guangzhou 510640,China) Abstract:Thedeterminationoftheanthocyaninscontentinblackriceoutlayerandthehumanmetaboliteswerestudied.After samplespreparation,theanthocyaninsweredeterminedwithanUV-HPLCsystemat520nmdetection.ACis-columnwasused atacolumntemperatureof300C.Themobilephase,4%aqueousH3POa/CH3CN(8.8:1.2V/V),wasusedunderisocraticconditions ataflowrateof0.8ml/min.Theresultsshowedthattotalanthocyanincontentsofblackriceoutlayerfractionwere2.31%. Cyanidin3-glucosideaccountedfor1.87%ofanthocyanincontentsandPeonidin3-glucosideaccountedfor0.44%ofanthocya- nincontents.TheplasmaCy-3-gmaxvaluewas21.47士4.48ng/mlat1.5hafterintakingtheblackriceoutlayerfractionandPn- 3-gcantbedetectedinplasmasample.TheurineCy-3-g-andPn-3-g-valueswere390.78士69.28ng/ml2hafterintake and105.90士26.26ng/ml4hafterintake,respectively.
