超声辅助提取蒲公英总黄酮及其抗氧化性研究.docx

黄酮类化合物作为植物次生代谢产物,因其抗氧化、抗炎等生物活性备受关注。蒲公英富含黄酮类物质,但其提取效率与活性受工艺条件限制。本研究以蒲公英为原料,采用超声辅助提取技术(UAE),系统优化超声时间、功率、温度、料液比及乙醇浓度对总黄酮提取率的影响,并通过DPPH自由基清除法对比其与合成抗氧化剂(VC、BHT)的活性差异。结果表明,最优工艺参数为超声时间40min、功率400W、温度60℃、料液比1:30g/mL、乙醇浓度80%,提取率达5.58%;抗氧化实验显示,蒲公英总黄酮在1.0mg/mL时DPPH清除率为74%,与BHT活性相当,且热稳定性与安全性显著优于合成抗氧化剂。研究证实了超声辅助提取的高效性及蒲公英黄酮的功能性价值,为其在食品、医药等领域的应用提供了理论支撑

Abstract
Flavonoids,assecondarymetabolitesinplants,haveattractedmuchattentionduetotheirbiologicalactivitiessuchasantioxidantandanti-inflammatoryproperties.Dandelionisrichinflavonoids,butitsextractionefficiencyandactivityarelimitedbyprocessconditions.Thisstudyuseddandelionastherawmaterialandemployedultrasoundassistedextractiontechnology(UAE)tosystematicallyoptimizetheeffectsofultrasoundtime,power,temperature,solid-liquidratio,andethanolconcentrationonthetotalflavonoidextractionrate.Theactivitydifferencesbetweendandelionandsyntheticantioxidants(VC,BHT)werecomparedbyDPPHradicalscavengingmethod.Theresultsshowedthattheoptimalprocessparameterswereultrasonictimeof40min,powerof400W,temperatureof60℃,solid-liquidratioof1:30g/mL,ethanolconcentrationof80%,andextractionrateof5.58%;TheantioxidantexperimentshowedthattheDPPHclearancerateoftotalflavonoidsindandelionwas74%at1.0mg/mL,whichwascomparabletoBHTactivity,andthethermalstabilityandsafetyweresignificantlybetterthansyntheticantioxidants.Thestudyconfirmedthehighefficiencyofultrasoundassistedextractionandthefunctionalvalueofdandelionflavonoids,providingtheoreticalsupportfortheirapplicationinfood,medicineandotherfields.
