胰岛素样生长因子-Ⅰ和其和髁突软骨发育调控[摘要]出生后的髁突软骨处于复杂的力学微环境中,其生长改建的生物学基础是髁突软骨细胞的增殖和分化。胰岛素样生长因子(IGF)-Ⅰ是髁突软骨生长发育过程中重要的生长因子之一。在细胞学水平,IGF-Ⅰ受胰岛素样生长因子连接蛋白(IGFBP)调控,通过与其受体的结合、激活来发挥生物学效应,其中包括调控细胞增殖、分化及程序性死亡,加快髁突软骨基质合成,参与髁突软骨细胞内力学信号的转导等。本文就IGF-Ⅰ及其受体,IGFBP及IGF-Ⅰ调控髁突软骨发育及其机制的研究作一综述。 [关键词]胰岛素样生长因子;髁突软骨;发育;作用机制 [中图分类号]Q 51[文献标志码]A[doi]10.3969/j.issn.1673-5749.2012.03.022 Insulin-like growth factor-Ⅰand its regulation on the development of mandibular condylar cartilage Jiang Liting, Gao Yiming.(Dept. of Stomatology, Ruijin Hospital, School of Medicine, Shanghai Jiao Tong University, Shanghai 200025, China) [Abstract]The postnatal condylar cartilage lives in complex mechanical micro-circumstances, and its growth and modification depends on proliferation and differentiation of condylar chondrocytes. Insulin-like growth factor(IGF)-Ⅰis one of the important growth factors during growth and development in the condylar cartilage. At the cellular level, IGF-Ⅰexerts its biological actions through the binding and activation of IGF-Ⅰ, and its bioefficacy is modulated by IGF binding proteins(IGFBP). IGF-Ⅰinduces variety cellular responses, including cell proliferation, differentiation, and aopotosis, promoting matrix synthesis and taking part in intercellular signal mechanotransduc-tion. The article reviews IGF-Ⅰand its receptor, IGFBP and its role and mechanism on the development of postnatal mandibular condylar cartilage. [Key words]insulin-like growth factor;condylar cartilage;development;role and mechanism 颞下颌关节应力敏感区的髁突软骨是下颌骨发育重要的调节位点,具有响应力学刺激并发生改建的能力[1]。下颌骨髁突的生长改建不仅受全身激素的控制,而且受胰岛素样生长因子(insulin-like growth factor,IGF)-Ⅰ等众多局部因素的影响[2-3]。IGF-Ⅰ是一种对关节软骨有自分泌调节作用的生长因子,在软骨、骨中质量丰富,对调控髁突软骨细胞的增殖活性、Ⅱ型胶原和蛋白聚糖的合成、细胞表型的维持[4]有着重要的意义,从而影响髁突软骨的生长发育。 1IGF-Ⅰ 1957年,Salmon等[5]发现了一种在体内起促生长作用,在体外试验中导致软骨细胞硫酸化的因子,他们认为,促生长素(growth hormone,GH)需要这个硫化因子为其效应中介,介导GH的合成代谢作用。1978年,这个硫化因子被正式命名为IGF。此后,有关IGF的结构、基因谱和生物学效应的研究[6-7]显示,IGF家族由IGF(IGF-Ⅰ、IGF-Ⅱ)、IGF受体(IGF-ⅠR、IGF-ⅡR)和IGF结合蛋白(insulin-like growth factor bindin
