第18 卷3 期 天津医科大学学报 Vol． 18, No． 3 2012 年9 月 Journal of Tianjin Medical University Sep． 2012 281 （ ） 文章编号 1006－8147 2012 03-0281-04 论 著 利用染色质免疫共沉淀测序研究Twist1 结合的 靶DNA 序列 1 1，2 1 1 1 1 1 1 孙涛 ，孙保存 ，赵秀兰 ，赵楠 ，古强 ，董学易 ，车娜 ，池嘉栋 （1. 天津医科大学病理学教研室，天津300070 ；2. 天津医科大学附属肿瘤医院病理科，天津300060） 摘要 目的：利用染色质免疫共沉淀测序研究Twist1 结合的靶DNA 序列，比较过表达Bcl-2 和正常状态下Twist1 结合靶DNA 序列的变化。方法：筛选常态表达Twist1 的肝癌细胞系，分别转染Bcl-2 表达质粒和对照空载体，收集转染后细胞以甲醛交联 DNA，收集样品进行染色质免疫共沉淀，获得与Twist1 蛋白结合的DNA 片段进行测序分析。结果：通过对5 种肝癌细胞系进行 筛选，观察到HepG2 可表达Twist1 ，其与DNA 结合序列在空载体对照组和Bcl-2 过表达组具有很大的差别，结合DNA 数量分 别为68 条和212 条，其中共享序列154 条。结论:Twist1 在不同环境下与DNA 结合位点多达212 个，涉及粘附分子、细胞骨架、 信号转导、代谢和转录调节等多种生物学功能。 关键词 Twist1 ；染色质免疫共沉淀；靶DNA 序列 中图分类号 R392.3 文献标志码 A Using chromatin immunoprecipitation to study the target DNA binding sequences of Twist1 1 1,2 1 1 1 1 1 1 SUN Tao , SUN Bao-cun , ZHAO Xiu-lan , ZHAO Nan , GU Qiang , DONG Xue-yi , CHE Na , CHI Jia-dong （1.
