恩施碎米荠叶片含硒蛋白的体外抗氧化研究
摘 要
硒在动物和人体生命活动中重要作用兼具营养、致毒、疾病治疗等多种生物学功能。恩施高硒区(恩施双河乡鱼塘坝)发现一种富硒能力极强的十字花科碎米荠属(Cardamine )植物碎米荠(Cardamine sp),建立了一个基于盐析和凝胶分离的蛋白质纯化流程;模拟机体产生超氧阴离子的体系研究了纯化的硒蛋白抗自由基的活性;以昆明小鼠为研究对象,探讨了纯化含硒蛋白的体外抗氧化活性研究:恩施碎米荠35%-55%饱和度硫酸铵沉淀硒蛋白具有清除自由基和抗脂质过氧化的能力,硒蛋白溶液的硒浓度为0.95 μg/mL时,对羟自由基和超氧阴离子的清除率超过50%, 含硒5.96 μg/mL的蛋白溶液对MDA生成的抑制率达到62.27%;在体外,对小鼠线粒体和肝脏MDA的生成具有明显的抑制作用,硒质量浓度为5.96 μg/mL的硒蛋白溶液可以保护红细胞免受过氧化损伤。叶片中的硒蛋白具有抗氧化活性

Abstract
selenium plays an important role in both nutrition, toxigenicity, disease treatment and so many kinds of biological function in animal and human life activity.The raw materials Cardamine is a plant of cruciferous Cardamine genera (Cardamine L) plants (Cardamine sp.) was found in Enshi high-selenium area (Yu tang ba in enshi double river township ) which has a strong se-enriched ability .established a separate based on salting-out and gel protein purification process, simulated a system of superoxide anion produced in body ,researched the anti-free radical activity of the selenium protein .To kunming mice as the research object, and discusseed the purification containing selenium protein in vitro antioxidant activity. Research showed that:the Soluble selenium protein in the Enshi Cardamine leaves has the ability of clearing the Supressing lipid peroxidation and Free radicals after it was precipitated by the Ammonium sulfate solution(35% - 55% ) ,when the concentration of the Soluble selenium protein is 0.95 μg/mL ,the rate of the cleared Hydroxyl radicals and superoxide anion is above 50%,the generated inhibition rate to the MDA reached 62.27% when Soluble selenium concentration is 5.96 μg/mL,under this rate,the red blood cells damage can be prevented from peroxidation at the same time.The Soluble selenium protein has a obvious inhibition to the mitochondria and liver MDA generation In vitro .Results showed that the Soluble selenium
