Introduction:
AuroraBkinaseisacrucialregulatorofmitosisandhasbeenimplicatedintumorigenesisandcancerprogression.Therefore,itisanattractivetargetforthedevelopmentofanticancerdrugs.Inrecentyears,anumberofsmallmoleculeinhibitorsofAuroraBkinasehavebeendeveloped,butmanyofthemsufferfrompoorpharmacokineticpropertiesandlimitedefficacy.Therefore,thereisaneedforthedevelopmentofnewagentswithimprovedefficacyandsafetyprofiles.
Objectives:
TheobjectivesofthisstudyaretosynthesizeaseriesofAuroraBkinaseinhibitorsusingacomputational-guideddesignapproach,andtoevaluatetheirbiologicalactivityinvitroandinvivo.
Methods:
Acomputational-guideddesignapproachwillbeusedtodesigntheAuroraBkinaseinhibitors.Thecompoundswillbesynthesizedusingstandardorganicsynthesistechniquessuchasnucleophilicsubstitution,Suzukicoupling,andSonogashiracoupling.ThepurityandstructureofthesynthesizedcompoundswillbeconfirmedusingNMRandmassspectrometry.TheAuroraBkinaseinhibitoryactivityofthecompoundswillbeevaluatedusinganinvitrokinaseassay.Themostpromisingcompoundswillbefurtherevaluatedincell-basedassaystoassesstheirantiproliferativeactivityagainstcancercells.Theselectivityandtoxicityofthecompoundswillalsobeevaluated.
ExpectedResults:
WeexpecttosynthesizeaseriesofpotentandselectiveAuroraBkinaseinhibitors.Thecompoundswillbecharacterizedbytheirchemicalstructuresandpurity.TheinvitrokinaseassayresultswillprovideinformationontheinhibitoryactivityofthecompoundsagainstAuroraBkinase.Thecell-basedassayswillprovideinformationontheantiproliferativeactivityofthecompoundsandtheirselectivity.Iftheresultsarepromising,wewillfurtherevaluatetheefficacyandtoxici
