谷氨酰胺联合5-氟尿嘧啶及环磷酰胺对胃癌细胞增殖影响[摘要] 目的:研究谷氨酰胺(Gln)联合5-氟尿嘧啶(5-FU)和环磷酰胺(CTX)对人胃癌SGC7901细胞周期和凋亡率的影响。方法:体外培养胃癌SGC7901细胞株,加入Gln及不同浓度的5-FU和CTX后收集细胞。应用流式细胞术检测胃癌细胞周期及凋亡率的变化。结果:5-FU加Gln时胃癌SGC7901细胞凋亡率不同,浓度由0 mg/L增加到240 mg/L时呈正直线相关(r=0.806,P0.05)。5-FU加Gln组G0/G1期细胞所占比例较CTX加Gln组明显增多(P0.05)。结论:Gln和5-FU影响胃癌细胞株的细胞周期和凋亡率。Gln和CTX对人胃癌细胞株的细胞周期和凋亡率均无明显影响。

Effect of combination of Glutamine (Gln) and 5-Fluorouracil (5-FU) and Cyclophosphamide (CTX) on the proliferation of human gastric carcinoma cells

[Abstract] Objective: To evaluate the effect of combination of Gln and 5-FU and CTX on the human gastric carcinoma cell line SGC7901 in vitro. Methods: Different concentrations of 5-FU or CTX and Gln were added in the cultured human gastric carcinoma cell line SGC7901 in vitro. The changes of apoptosis rate and cell cycle were detected by the flow cytometry analysis. Results: Combination of Gln and 5-FU could inhibit the cell growth and enhanced apoptosis rate. The concentration of 5-FU and the apoptosis rate of gastric carcinoma cell line SGC7901 had positive linear correlation (r=0.806, P0.05). The rate of G0/G1 phase in cell cycle of 5-FU and Gln group was more than that of CTX and Gln group (P0.05). Conclusion: Combination of Gln and 5-FU can inhibit the cell growth and enhance apoptosis rate. There is no change of apoptosis rate and cell cycle by combination of CTX and Gln.

我国是胃癌高发国家之一,发病人数约占世界总发病人数的35%,每年有超过26万人死于胃癌,占所有癌症死亡人数的1/5[1]。目前在胃癌治疗中,手术、化疗仍是主要手段。改善胃癌患者的营养状况和免疫功能,减少化疗药物的毒副作用显得尤为重要[2]。谷氨酰胺(Gln)是各种细胞生长所必需的物质,能促进机体细胞生长和蛋白质合成,增强免疫细胞功能。本实验探讨Gln联合化疗药物5-氟尿嘧啶(5-FU)和环磷酰胺(CTX)对胃癌细胞增殖的影响。
