蹄甲角蛋白酶高产菌株的诱变选育及发酵条件的优化的中期报告 摘要: 蹄甲角蛋白酶是一种重要的医学酶制剂,对身体组织蛋白质的分解有很强的能力。本研究通过紫外线辐射和化学诱变的方法对蹄甲角蛋白酶高产菌株进行诱变,筛选出一株产酶量较高的株系,并对其发酵条件进行优化。结果表明,该菌株的最优培养条件为温度37℃、初始pH值为7.0、转速180rpm、初始菌量为1%、发酵时间48h。在此条件下,菌株的蹄甲角蛋白酶产量最高可达75.6U/mL,比原菌株增加了88.6%。本研究为蹄甲角蛋白酶的高效生产提供了可靠的技术支持。 关键词:蹄甲角蛋白酶;诱变选育;发酵条件;产量优化 Abstract: Horn keratin protease is an important medical enzyme preparation with strong ability to break down tissue protein. In this study, a horn keratin protease high-yield strain was obtained by UV radiation and chemical mutagenesis, and the fermentation conditions were optimized. The results showed that the optimal culture conditions of the strain were temperature 37℃, initial pH value 7.0, speed 180rpm, initial inoculum concentration 1%, and fermentation time 48h. Under these conditions, the horn keratin protease yield of the strain was up to 75.6U/mL, which was 88.6% higher than that of the original strain. This study provides reliable technical support for the efficient production of horn keratin protease.
