Nucleic acid sensors and SLEAbstract:Systemic lupus erythematosus (SLE) is a chronic autoimmune multisystemic disease, which is characterized by deficiency in clearance of self nuclear antigens.Studies have found nucleic acid sensors take part in the pathogenesis and development of SLE.Therefore, this article will provide an integral and comprehensive review of nucleic acid sensors associated with SLE, focusing on sorts of nucleic acid sensors, signaling pathway and the immune response triggered by them. What’s more, therapy targeting nucleic acid sensors will be discussed as well.Key words: SLE, DNA sensor, RNA sensorIntroduction:1.SLE:Systemic lupus erythematosus (SLE) is a potentially fatal disease, which affects multiple organs including the skin, joints, lung, kidney, and brain that eventually leads to organ failure and premature death[1-4]. The precise etiology of SLE remains elusive,however;inefficient clearance of self nuclear antigens released from normal or pathological cell death from bloodstream has been reported as an important factor leading to the initiation and development of SLE?[5-7].?Over recent years, nucleicsensors for DNA and RNAhave been found as novel partof innate immune. Toll-like receptor (TLR) 9, retinoic acid-inducible gene I (RIG-I)-like receptors (RLRs) including RIG-I, melanoma differentiation-associated gene-5 (MDA5), DNA-dependent RNA polymerase III (Pol-III), DNA binding protein 1 (ZBP1) or DNA-dependent activator of IFN-regulatory factor (DAI), IFN-inducible p200-protein including, absent in melanoma 2 (AIM2), interferon-inducible protein 16 (IFI16) have been identified as DNA sensor[8-12]. What’s more, DExDc helicase DDX41, DHX36 and DHX9, Leucine-rich repeat flightless-interacting protein 1 (LRRFIP1), Ku70 were recently found to be novel DNA sensors[13-16].
