中草药 Chinese Traditional and Herbal Drugs 第 44 卷 第 4 期 2013 年 2 月 • 449 • 蜜环菌菌索多糖延缓秀丽隐杆线虫衰老机制研究 1 1 1* 2 陈亮稳 ,张云侠 ,于 敏 ,王顺昌 1. 安徽大学生命科学学院 安徽大学中药研究与开发重点实验室,安徽 合肥 230039 2. 淮南师范学院 生命科学系,安徽 淮南 232000 摘 要:目的 探讨蜜环菌菌索多糖(AMP )延缓秀丽隐杆线虫衰老的作用机制。方法 以模式生物秀丽隐杆线虫作为活 体模型,测定其在正常培养条件下的生存期与后代平均数量,氧化应激条件下的存活率和热休克蛋白 16.2 (HSP-16.2 )、超 氧化物歧化酶-3 (SOD-3 )的表达。结果 在正常培养条件下,蜜环菌菌索多糖(AMP-1 、AMP-2 )显著延长秀丽隐杆线虫 的生存期,对其生殖力没有损害;在氧化应激条件下,显著提高秀丽隐杆线虫HSP-16.2 和 SOD-3 的表达。结论 提高秀丽 隐杆线虫的氧化应激能力可能是AMP 延缓秀丽隐杆线虫衰老的作用机制。 关键词:蜜环菌菌索多糖;抗衰老;秀丽隐杆线虫;热休克蛋白;超氧化物歧化酶-3 中图分类号:R282.710.5 ;R977.9 文献标志码:A 文章编号:0253 - 2670(2013)04 - 0449 - 05 DOI: 10.7501/j.issn.0253-2670.2013.04.015 Anti-aging mechanism of polysaccharide from rhizomorph of Armillaria mellea in Caenorhabditis elegans 1 1 1 2 CHEN Liang-wen , ZHANG Yun-xia , YU Min , WANG Shun-chang 1. Methods With C. elegans as living model, the life span and the average number of offsprings were determined under normal culture conditions; The survival rate under oxidative stress and the expression of heat shock protein-16.2 (HSP-16.2) and superoxide dismutase-3 (SOD-3) were determined.
