Introduction:
TRPM7isanon-selectivecationchannelthatplaysanimportantroleintheregulationofintracellularcalciumandmagnesiumhomeostasis,aswellasinthephysiologicalfunctionsofavarietyoftissues,includingthecardiovascularsystem.StudieshaveshownthatTRPM7ishighlyexpressedincardiacmyocytesandisinvolvedintheregulationofcardiacexcitabilityandcontractility.OverexpressionofTRPM7hasbeenlinkedtocardiachypertrophyandheartfailure,whileitsdownregulationhasbeenshowntoimprovecardiacfunctioninanimalmodels.However,theprecisemechanismsunderlyingtheseeffectsarenotfullyunderstood.
Objectives:
TheaimofthisstudyistoinvestigatethefunctionalconsequencesofTRPM7downregulationinratcardiacmyocytesandtoexplorethepossiblemechanismsinvolved.
Methods:
Ratcardiacmyocyteswillbeisolatedandculturedinvitro.TheexpressionlevelofTRPM7willbedownregulatedusingsmallinterferingRNAtechnology.TheeffectsofTRPM7downregulationoncardiacfunctionwillbeevaluatedbymeasuringtheactionpotentialduration,calciumtransients,andcontractilityparametersofthemyocytes.Theexpressionandphosphorylationlevelsofkeyregulatoryproteinsinvolvedincardiacexcitation-contractioncouplingandhypertrophysignalingpathwayswillalsobeexamined.
ExpectedResults:
WeexpecttoobserveanimprovementincardiacfunctionfollowingTRPM7downregulation,asevidencedbyincreasedcalciumhandlingandcontractilityparameters.Wealsoexpecttoseeadecreaseinthephosphorylationlevelsofkeyhypertrophysignalingproteins,suggestingthatTRPM7downregulationmaypreventcardiachypertrophyandheartfailure.
Significance:
