异氟醚对大鼠空间记忆及海马中ApoE(载脂蛋白e)表达的影响

【摘要】 目的 研究异氟醚对大鼠空间记忆及海马组织中载脂蛋白e(ApoE)阳性细胞表达的影响。方法 12月龄雄性wistar(350g)大鼠48只,随机分为三组(n=16),对照组(不麻醉,吸入30%氧气的空氧混合气体),A组(异氟醚1.5%,连续吸入5d,每天3h),B组(异氟醚2%,连续吸入5d,每天2h)。各组大鼠在停药后1d取8只行Morris水迷宫进行认知功能测试,其余处死取海马组织,用免疫组织化学方法测定ApoE的表达。结果 各组大鼠异氟醚吸入过程中生命体征平稳,血氧饱和度及心率均在正常范围。A组和B组逃避潜伏期和游泳距离明显长于对照组(Plt;0.05),ApoE在A组和B组的表达明显高于对照组(Plt;0.05)。结论 多次吸入异氟醚对大鼠的认知功能有所改变,可能与海马组织ApoE的表达有关。

Effects of Isoflurane on spatial memory and expression of ApoE (Apolipoprotein e) in hippocampus in rats
【Abstract】 Objective To study isoflurane on spatial memory and the expression of apolipoprotein e (ApoE) positive cells in hippocampus in rats. Method 48 male wistar rats12-month-old (350g) Were randomly divided into three groups (n = 16),the control group (no anesthesia, isoflurane of 30% oxygen empty oxygen mixed gas),group A (isoflurane 1.5% continuous inhalation 5d, 3h daily),group B (continuous inhalation of isoflurane 2% for 5d, 2h daily). Eight rats were tested cognitive function by the Morris water maze test in each group one day after stopping inhalation, the rest were sacrificed to measure the expression of ApoE in hippocampus by immunohistochemical methods. Result The vital signs of rats in each group were stable, oxygen saturation and heart rate were within the normal range when isoflurane inhalation.The escape latency and swimming distance was significantly longer in Group A and Group B than that of the control group (Plt;0.05) The expression of ApoE was significantly higher in group A and group B(Plt;0.05). conclusion Changes in cognitive function in rats repeatedly inhaled isoflurane may ApoE expression and hippocampus.
