雷公藤内酯醇对阿尔茨海默病模型大鼠海马白细胞介素1β mRNA和蛋白表达的影响 (作者:___________单位: ___________邮编: ___________) 作者:吕诚 胡小令 杨宝林 万斌 聂菁 薛国勇 【摘要】   目的 探讨雷公藤内酯醇对阿尔茨海默病(AD)模型大鼠海马白细胞介素1β(IL1β)mRNA和蛋白表达的影响。方法 30只SD雄性大鼠等分成对照组、模型组、用药组。模型组大鼠给予双侧海马各一次性注射凝聚态Aβ140,对照组大鼠海马注射等量生理盐水,用药组大鼠在海马注射凝聚态Aβ140后腹腔注射雷公藤内酯醇,RTPCR和免疫组织化学方法检测各组大鼠海马IL1βmRNA和蛋白的表达。结果 免疫组织化学染色显示,模型组IL1β阳性细胞数和平均光密度明显高于对照组(P<0.01),用药组IL1β阳性细胞数和平均光密度较模型组明显减少(P0.05或0.01)。RTPCR结果显示,模型组IL1βmRNA水平明显高于对照组(P<0.01),用药组IL1βmRNA水平较模型组明显下降(P<0.01)。结论 雷公藤内酯醇对AD模型大鼠海马IL1β的表达有抑制作用。 【关键词】 雷公藤内酯醇;阿尔茨海默病;白细胞介素1β;海马 【Abstract】 Objective To explore the effects of triptolide(TP) on the expression of interleukin1β mRNA and protein of hippocampus in model rats with Alzheimer′s disease (AD). Methods Thirty male SD rats were equally divided into control,AD model and TPtreated groups. The AD model group was made with bilateral microinjection of aggregated betaamyloid protein(Aβ)140 into hippocampus in rats and the control group rats were injected with normal saline in the same way.The TPtreated group rats were administered TP intraperitoneally after microinjection of aggregated Aβ140 into hippocampus. The expressions of interleukin1β mRNA and protein of hippocampus in each group were assayed by reverse transcription polymerase chain reaction(RTPCR) and immunohistochemical staining. Results The cell number and average optical density of IL1β positive staining in the AD model group were higher than those in the control group(P<0.01), the cell number and average optical density of IL1βpositive staining in the TPtreated group were lower than those in the AD model group(P<0.05 or 0.01). The expressions of IL1βmRNA in the AD model group was higher than that in the control group(P<0.01). The expression of IL1βmRNA in the TPtreated group was lower than that in the AD model group(P<0.01).Conclusions TP can inhibit IL1β expression of hippocampus in model rats with AD. 【Key words】 Triptolide; Alzheimer′s disease; Interleukin1β; Hippocamp
