雷公藤内酯醇对阿尔茨海默病模型大鼠海马白细胞介素1β mRNA和蛋白表达的影响

【摘要】 目的 探讨雷公藤内酯醇对阿尔茨海默病(AD)模型大鼠海马白细胞介素1β(IL1β)mRNA和蛋白表达的影响。方法 30只SD雄性大鼠等分成对照组、模型组、用药组。模型组大鼠给予双侧海马各一次性注射凝聚态Aβ140,对照组大鼠海马注射等量生理盐水,用药组大鼠在海马注射凝聚态Aβ140后腹腔注射雷公藤内酯醇,RTPCR和免疫组织化学方法检测各组大鼠海马IL1βmRNA和蛋白的表达。结果 免疫组织化学染色显示,模型组IL1β阳性细胞数和平均光密度明显高于对照组(P<0.01),用药组IL1β阳性细胞数和平均光密度较模型组明显减少(P0.05或0.01)。RTPCR结果显示,模型组IL1βmRNA水平明显高于对照组(P<0.01),用药组IL1βmRNA水平较模型组明显下降(P<0.01)。结论 雷公藤内酯醇对AD模型大鼠海马IL1β的表达有抑制作用。

【Abstract】 Objective To explore the effects of triptolide(TP) on the expression of interleukin1β mRNA and protein of hippocampus in model rats with Alzheimer′s disease (AD). Methods Thirty male SD rats were equally divided into control,AD model and TPtreated groups. The AD model group was made with bilateral microinjection of aggregated betaamyloid protein(Aβ)140 into hippocampus in rats and the control group rats were injected with normal saline in the same way.The TPtreated group rats were administered TP intraperitoneally after microinjection of aggregated Aβ140 into hippocampus. The expressions of interleukin1β mRNA and protein of hippocampus in each group were assayed by reverse transcription polymerase chain reaction(RTPCR) and immunohistochemical staining. Results The cell number and average optical density of IL1β positive staining in the AD model group were higher than those in the control group(P<0.01), the cell number and average optical density of IL1βpositive staining in the TPtreated group were lower than those in the AD model group(P<0.05 or 0.01). The expressions of IL1βmRNA in the AD model group was higher than that in the control group(P<0.01). The expression of IL1βmRNA in the TPtreated group was lower than that in the AD model group(P<0.01).Conclusions TP can inhibit IL1β expression of hippocampus in model rats with AD.
