利用糖蜜发酵生产丁醇菌株的分离、筛选及鉴定

:高效糖蜜丁醇的菌株本从自然(牛粪,沼气池,土壤)采样通过玉米培养基富集初筛分离获得12株生产丁醇能力较强的菌株。用8糖蜜发酵复筛有2株菌株丁醇产量较高,分别为8.57 g/L和8.45 g/L,丁醇比高达75 %。将菌株分别命名为gxzp-5和gxzp-9,然后对这两株发酵性能优良的菌株进行了分子鉴定,经过16S rDNA基因序列同源性分析初步鉴定都为拜氏梭菌(Clostridium beijerinckii)。两菌株为下一步的菌种基因改造工作提供了研究基础,也为国内拜氏梭菌的工业应用及基础研究奠定了理论基础。

Abstract: In order to obtain efficient molasses-fermentation strains, samples were got from cow dung, methanetank and soil,and then 12 strains were screened by enrichment on corn medium,which can effectively produce butanol. Two strains re-screened by 8BX molasses can convert molasses into butanol more effectively,the yield of which is 8.57 g/L and 8.45 g/L respectively,and the rate of butanol/total solvent reached 75 %. These two strains were named gxzp-5 and gxzp-9. They were identified as Clostridium beijerinckii through 16S rDNA sequence analysis,which could be used not only as orginal slrains for further genic alteration work,but also as theory base for industrial application and basic research of Clostridium beijerinckii in our country .

随着石化资源的枯竭,石油价格的不断攀升,微生物发酵生产丁醇受到了人们的普遍关注。丁醇主要溶剂、抗氧剂、增塑剂、合成药物精细化工在能源危机的今天,由于丁醇能量密度高,亲水性低,蒸汽压低,辛烷值高等特点,被认为最有发展潜力的新型液体燃料之一。基金项目:桂科攻0992022-1,桂科广西大学开放课题J0804,科学院基本业务费09YJ17SW05
