Med J Chin PLA, Vol. 39, No. 1, January 1, 2014  25 论 著 青蒿琥酯对食管癌Ec9706细胞线粒体膜电位及凋亡的影响 刘亮,左连富,王静 [摘要] 目的 研究青蒿琥酯(Art)对食管癌Ec9706 细胞线粒体膜电位的影响,探讨Art 诱导食管癌Ec9706 细胞凋 亡的作用机制。方法 以不同浓度(30、60 、120μmol/L) 的Art 作用于食管鳞癌Ec9706 细胞12h ,对照组以生理盐水代替 Art 。光学显微镜下观察细胞形态变化,采用流式细胞术Annexin V/PI 双染法检测细胞凋亡水平,荧光染料罗丹明123 染色检测细胞线粒体膜电位水平。采用流式细胞术检测Caspase-3蛋白表达水平。结果 Art 作用后Ec9706 细胞形态上 呈现不同程度的变化,贴壁细胞变圆,体积变小,飘浮于细胞培养液中。不同浓度Art 作用12h后,Ec9706 细胞表现出 不同程度的细胞凋亡且呈药物剂量依赖性,与对照组相比差异有统计学意义(P 0.01) ;Ec9706 细胞线粒体膜电位表现 出不同程度降低且呈药物剂量依赖性,与对照组相比差异有统计学意义(P 0.05) 。与对照组相比,Art 组Ec9706 细胞中 Caspase-3蛋白表达水平显著增加(P 0.01) ,且呈药物剂量依赖性。结论 Art诱导Ec9706 细胞凋亡的机制可能是通过降 低细胞线粒体膜电位,启动内源性线粒体凋亡途径,激活Caspase-3蛋白,从而诱导细胞凋亡。 [关键词] 青蒿琥酯;细胞凋亡;膜电位,线粒体;流式细胞术 [中图分类号] R735.1    [文献标志码] A    [文章编号] 0577-7402(2014)01-0025-05 [DOI] 10.11855/j.issn.0577-7402.2014.01.06 Effects of artesunate on membrane potential of mitochondria and cell apoptosis of Ec9706 cells * LIU Liang, ZUO Lian-Fu , WANG Jing Department of F CM A nalysis, Tumor Institute, The Fourth Hospital of Hebei Medical University, Shijiazhuang 050011, China * Corresponding author, E-mail: zuolianfu4909@ This work was supported by the Natural Science Foundation of Hebei Province (H2012206107); M edical Science R esearch Project of Hebei P rovince ; Construction Foundation f or the Subject of Oncology in the Universities of Hebei P rovince[Ji High- Teach(2005)No.52] [Abstract] Objective To explore the apoptotic mechanism of esophageal cancer Ec9706 cells induced by artesunate (Art) by means of observing the effect of Art on the membrane potential of Ec9706 cell mitochondria.
