What we have heard from Chris has been an insight into an important part of the history of modern biotechnology, and the story of one of its enabling tools, FPLC System. The role of this system can be understood from Dr. Fullers presentation, and is highlighted by the fact that it has been chosen for display here in this exhibition of modern biochemistry. After this look into history, a history still being made today, I would now like to bring you a glimpse of the future in protein purification and present the most recent addition to our ?KTAdesign line of chromatography systems. ?KTA is a swedish word which means real, genuine or true. ?KTAdesign is also the name of a platform of chromatography systems from Amersham Pharmacia Biotech. Introduced in 1996, ?KTAdesign stands for a new way of working in biomolecule purification. * HeartJust like a human heart, the pump of the system has to be extremely reliable. The gradient pump of ?KTA FPLC improves on the reliable technology used in FPLC System, but can operate at up to 20 ml/min, more than twice the flow rate. The signicance of this is not primarily to do two-minute runs, but to be able to run large high capacity columns early in a purification scheme. The pump is insensitive to high salt concentrations and air. Eyes and NoseIf the pump is the heart of the system, the monitor is its senses, its eyes and nose. The monitor includes sensitive UV detection to detect proteins as they elute, with automatic scaling to avoid the detector going out of range, a common problem in protein work. Conductivity detection monitors elution conditions. It is fully prepared for pH monitoring as well. BrainThe brain of ?KTA FPLC is UNICORN 3.0 for Windows NT. This is the software as in other ?KTAdesign systems, now transferred to the Windows NT operating system, which will allow for even easier networking.
