广东农业科学 2014 年第 9 期 155 一株源于红树林土壤芽孢杆菌的鉴定 及其蛋白酶学性质研究 谢为天 李红胜 刘 颖 徐春厚 广东海洋大学农学院袁广东 湛江 524088冤 摘要 从湛江红树林土壤中分离出一株产蛋白酶的芽孢杆菌袁通过细菌形态观察尧生理生化试验及 16S rRNA 基因序列进行分类鉴定袁并研究了温度尧pH 值尧金属离子尧EDTA 和吐温 80 等因素对该菌株发酵产蛋白酶的酶学性 质的影响遥 结果表明袁该菌株为解淀粉芽孢杆菌袁命名为解淀粉芽孢杆菌 y-6 Bac illus amy loliq uefac ie ns strain y -6冤曰 该菌株产生蛋白酶的最适温度为 55益袁最适 pH 为 7.0 ~7.5 袁40益时袁酶活性稳定袁保温 3 h 仍有 95% 以上的相对酶活 力袁pH 为 5.5~11 时袁酶活性稳定袁相对酶活力均在 90% 以上曰Ca2+尧Zn2+和吐温 80 对蛋白酶有一定的激活作用袁Mn2+ 尧 Fe3+尧Cu2+及 EDTA 对酶有一定的抑制作用袁其中 EDTA 对酶的抑制作用最强遥 关键词 芽孢杆菌曰 鉴定曰 蛋白酶曰 酶学性质曰 红树林 中图分类号 Q814 文献标识码 A 文章编号 员园园源原愿苑源载渊圆园员源冤园怨原0155-04 Study on identification and protease properties of a bacillus isolated from mangrove soil XIE Wei-tian, LI Hong-sheng, LIU Ying, XU Chun-hou (A gric ultural Colle ge , G uangdong Oce an Unive rs ity , Zhanj iang 524088, China) Abstract: A strain of bacillus w hich could produce protease was isolated from soil sample collected from a chicken farm, and was identified by the observation of its morphology, culture characteristics and the analysis of 16S rRNA sequence. T he properties of protease excreted by isolate fermentation were studied w ith different temperature, pH, metal ion, EDTA and Tween 80. Results showed that, the isolate belonged to Bac illus amyloliq uefac ie ns, and was named Bac illus amyloliq ue fac ie ns strain Y -6, the optimal reaction temperature of protease produced by strain Y -6 was 55益, the optimal reaction pH was 7.0 ~7.5.
