N-羧甲基壳聚糖对皮肤创伤修复功能的研究 摘要 甲壳素(Chitin)是自然界可再生的一种聚乙酰氨基葡萄糖高分子多糖。壳聚糖(Chitosan)是甲壳素脱乙酰基的产物,由于其资源丰富、天然无毒、无抗原性、生物相容性好、可生物降解且具有特殊的生物活性,近10多年来,对壳聚糖及其衍生物的性质、功能的研究和应用开发已得到广泛的重视。其研究应用范围已涉及到工业、农业、生物工程、医药卫生、食品工业、生物医用材料、畜牧水产、化学化工等众多领域。在医学领域更是取得了长足的进展,已有较多报道。甲壳素和壳聚糖均不能直接溶于水,极大的限制了应用。而壳聚糖C6位羟基最活泼易于进行羧化反应,通过改性后的羧甲基壳聚糖(CM-Chitosan)具有良好的水溶性,因此在医药、化妆品等领域有着广泛的应用前景。 皮肤创面组织正常而又快速的修复一直是基础医学的研究热点之一,壳聚糖及其衍生物具备的诸多优良性质使其成为该领域颇受关注的天然高分子材料 本研究拟在多年壳聚糖相关研究基础上,对目前尚未报道的N-羧甲基壳聚糖(N-CMC)对创伤修复的作用和机理进行研究。首先以壳聚糖、乙醛酸为原料,制备N-CMC敷料,对其羧化度、脱乙酰度、分子结构、生物相容性等性质进行评价;继而制备Ⅱ度烫伤动物模型,根据创面出血量、愈合创面局部特征、创面愈合率、创面愈合时间、组织学特征等指标评价创面修复效果 关键词 N-CMC、羧化度、水溶性、创伤修复 Thesis Title Chitin (Chitin) is a natural renewable polymer poly acetylglucosamine polysaccharide. Chitosan (Chitosan) is the product of chitin deacetylation, because of its abundant resources, natural non-toxic, non-antigenic, biocompatible, and can be biodegradable and has a specific biological activity of the past 10 years, on the nature of chitosan and its derivatives, function and application development has been widely appreciated. Application of their research has been involved in industry, agriculture, biological engineering, medicine and health, food industry, biomedical materials, animal aquaculture, chemical industry and other fields. In medicine is made remarkable progress, there are more reports. Chitin and chitosan are not directly soluble in water, which greatly limits the application. The C6 hydroxyl groups of chitosan were the most lively and easy-to-carboxylation reaction, through the modified carboxymethyl chitosan (CM-Chitosan) with good water-soluble, so in medicine, cosmetics and other fields have broad application prospects.
