8羟基脱氧鸟苷在2型糖尿病大鼠肾脏表达及瑞舒伐他汀钙影响【摘要】 目的 观察8-羟基脱氧鸟苷(8-OHdG)在2型糖尿病模型大鼠肾脏的表达及瑞舒伐他汀钙(Rosuvastatin)对它的影响。方法 将大鼠分为正常对照组(C)、糖尿病组(D)、瑞舒伐他汀钙治疗组(N),比较各组大鼠肾组织中丙二醛(MDA)含量、超氧化物岐化酶(SOD)活性,和8羟基脱氧鸟苷(8-OHdG)含量,同时观察血糖、血胰岛素、肾功能和肾脏形态变化。结果 与正常组比较,糖尿病组血液中MDA含量明显上升(P
1.2.2.2 肾组织中MDA含量、SOD活性的测定 取0.5 ml组织匀浆,低温离心,取上清液,采用化学比色法测定光吸收度,计算MDA、SOD含量。?
1.2.2.3 肾组织免疫学检查 常规脱蜡,封闭,一抗37℃孵育2 h,4℃冰箱过夜,标记二抗,作免疫染色,应用高清晰度彩色免疫图像分析系统(IPP55.0)分析,测定肾小球(平均光密度)直径。?
1.2.2.4 肾脏组织电镜观察 肾组织固定,脱水,包埋,切片,染色,在Hitachi-7500透射电镜下观察。?
1.3 统计学方法
