精品论文 参考文献 糖尿病大鼠肾脏解耦连蛋白2与氧化应激的关系 崔国峰1 徐建伟1 王雁飞2   1青岛市第八人民医院肾脏内科 266100 2青岛大学附属医院肾脏内科266000   【摘要】目的 研究解耦连蛋白2(UCP2)在糖尿病大鼠肾脏中的表达,探讨线粒体氧化应激水平与UCP2表达的相关意义。方法 链脲佐菌素60mg/kg腹腔注射构建1型糖尿病大鼠模型,造模成功者为糖尿病组(D组 n=25),并设置健康对照组(C组 n=25)。结果 D组肾脏的病理改变于4周时变化不明显,8周及12周较C组明显加重;Western-blot显示D组4周、8周及12周肾脏中UCP2的表达逐渐升高,且明显高于同时间的C组(Plt;0.05)。结论 UCP2在糖尿病大鼠肾脏中表达的升高,表明线粒体氧化应激在糖尿病肾病的发展中起重要作用。   【关键词】糖尿病肾病;氧化应激;UCP2   The relationship of UCP2 and oxidative stress in the kidney of diabetic rats   Cui Guofeng1, Xu Jianwei1,Wang Yanfei2   1Qingdao Eighth Peoples Hospital of renal Department of Internal Medicine 266100   2Department of internal medicine, Affiliated Hospital of Qiingdao University 266000   【Abstract】Objective To investigate the expression of UCP2 and its mechanism with oxidative stress in the kidney of STZ induced diabetic rats. Method 50 rats were randomly divided into two groups: control group (group C n=25)and diabetic group (group D n=25).Rats in group D were given STZ (60mg/kg) by intraperitoneal injection to establish diabetic rat model. Result The pathological changes of the kidney in group D were not distinct at 4th weeks, but the pathological changes were much more serious than group C at 8th weeks and 12th weeks. The expression of UCP2 in group D were much higher than group C at 4th、8th and 12th weeks in Western-blot, the differences had statistical significance (Plt;0.05).Conclusion The expression of UCP2 increases in diabetic rat model, which indicates that mitochondrial may take part in the progression of diabetic nephropathy.   【Key words】diabetic nephropathy;oxidative stress;UCP2   目前随着生活质量的提高及饮食结构的改变糖尿病的发病率逐年升高,我国糖尿病的发病率位居世界第二,糖尿病肾病是糖尿病较严重的并发症之一,并且在我国已成为继慢性肾小球肾炎之外的引起终末期肾病的主要原因,部分西方国家糖尿病肾病则是引起终末期肾病的首要原因。目前大量研究表明氧化应激在糖尿病肾病的发生及发展中起重要作用。UCP2是解耦连蛋白家族中的成员,位于线粒体的内膜上,可以使呼吸链中氧化与磷酸化解耦连,从而减少活性氧产物(ROS)的产生。但目前为止尚未见关于UCP2在糖尿病肾脏中的表达。本实验通过研究糖尿病大鼠肾脏中UCP2的表达情况,评估线粒体的氧化应激水平,进一步探讨糖尿病肾病的发病机制。   一、材料与方法   1. 实验动物与主要试剂   6-8周清洁级,体质量为220-250g健康雄性Wistar大鼠,购自山东鲁抗实验动物中心,兔抗大鼠UCP2抗体(美国S
