摘要:
本文旨在研究转PEPC基因水稻在干旱逆境下的生理反应。通过小麦胚芽粉肪酸和PEG(聚乙二醇)溶液处理,筛选出抗干旱能力较强的材料。接着,利用PCR和RT-PCR技术鉴定转入PEPC基因的水稻植株,确认其基因组中PEPC基因的存在,并检测其表达水平。然后,将转PEPC基因的水稻植株和野生型水稻植株分别置于干旱胁迫模拟器中进行处理,通过比较两者在干旱逆境下的生理指标,如叶片水势、叶绿素含量、可溶性糖和脯氨酸等,分析转PEPC基因水稻对干旱逆境的响应和适应机制。最后,通过分析PEPC基因对水稻干旱逆境的调控作用,探索其应用于作物抗旱育种的潜力。

Abstract:
The aim of this study is to investigate the physiological response of transgenic rice overexpressing the PEPC gene under drought stress. The materials with strong drought tolerance will be screened by treating wheat germ powder with fatty acids and PEG (polyethylene glycol) solution. Then, PCR and RT-PCR techniques will be used to identify the presence and expression level of the PEPC gene in rice. Afterwards, the transgenic rice plants and wild-type rice plants will be subjected to drought stress simulation, and the physiological indicators of both groups, such as leaf water potential, chlorophyll content, soluble sugar and proline, will be compared to analyze the response and adaptation mechanism of transgenic rice to drought stress. Finally, the regulatory effect of the PEPC gene on drought stress in rice will be analyzed to explore its potential for drought-resistant breeding.
