进出口食品中不同血清型沙门氏菌 PFGE 和MLST 分型比较研究刘慧 进出口食品中不同血清型沙门氏菌 PFGE 和 MLST 分型比较研究 刘慧玲 1, 万志刚 1, 洪小柳 1, 谢丹妮 1, 黄李华 1, 赵 芳 1, 薛 峰 2, 蒋 源 2, 吕敬章 1* (1. 深圳出入境检验检疫局食品检验检疫技术中心, 深圳市食品安全检测技术研发重点实验室, 深圳 518045; 2. 江苏出入境检验检疫局动植物与食品检测中心, 南京 210001) 摘 要: 目的 通过脉冲场凝胶电泳(PFGE)和多位点序列分型(MLST)两种方法对进出口食品中不同血清型沙 门氏菌的分辨力和应用价值进行了比较研究。方法 采用 PFGE 和 MLST 两种方法对进出口食品中分离得到 的鼠伤寒及肠炎两种血清型共 30 株沙门氏菌进行分型研究。结果 两种血清型共 30 株沙门氏菌经 PFGE 分 型可得到 23 种型别, DI 值为 0.9563; 鼠伤寒和肠炎两种血清型菌分别经 PFGE 分型, DI 值分别为 1.000 和 0.9048。MLST 对 30 株沙门氏菌分型得到 4 种 ST 型, DI 值为 0.5793; 鼠伤寒和肠炎两种血清型菌分别经 MLST 分型, DI 值分别为 0.2571 和 0.1333。结论 在分辨力方面, PFGE 更适合于同一沙门氏菌血清型的分型, MLST 则适合于不同沙门氏菌血清型间的分型。MLST 在替代、补充血清型分型方面有潜在的优势, PFGE 在溯源研 究方面更胜一筹。 关键词: 食品; 鼠伤寒沙门氏菌; 肠炎沙门氏菌; 脉冲场凝胶电泳; 多位点序列分型 Comparison of pulsed-field gel electrophoresis and multilocus sequence typing of Salmonella in import and export foods LIU Hui-Ling1, WAN Zhi-Gang1, HONG Xiao-Liu1, XIE Dan-Ni1, HUANG Li-Hua1, ZHAO Fang1, XUE Feng2, JIANG Yuan2, LV Jing-Zhang1* (1. Methods All 30 strains of both Salmonella typhimu- rium and Salmonella enteritidis isolates which were separated from import and export foods were typed by PFGE and MLST. Results All 30 strains Salmonella were divided into 23 PFGE types with discriminatory index 0.9563 while they were divided into 4 ST types by MLST with discriminatory index 0.5793. The discri- minatory index of Salmonella typhimurium and Salmonella enteritidis isolates typed by PFGE were 1.000 and 0.9048 with the contrast of 0.2571 and 0.1333 by MLST. Conclus
