安徽农业大学学报, 2016, 43(4): 593-597 Journal of Anhui Agricultural University [DOI] 10.13610/ki.1672-352x009 网络出版时间:2016-7-12 16:03:35 [URL] /kcms/detail/34.1162.S1603.018.html 来自安徽不同地区黄瓜绿斑驳花叶病毒 (CGMMV )cp 基因的分子进化分析 李 帅,单文书,冯明峰,蒋西子,江 彤* (安徽农业大学植物保护学院,合肥 230036) 摘 要:为了揭示黄瓜绿斑驳花叶病毒(Cucumber green mottle mosaic virus , CGMMV )安徽分离物分子变异 及系统进化情况,从安徽省5 个地区采集感染CGMMV 的葫芦科样本。提取感病样本总RNA ,经RT-PCR 扩增、 克隆和测序,获得 17 个CGMMV 分离物的cp 基因序列。序列比对发现,17 个CGMMV 安徽分离物的cp 基因核 苷酸序列相似性极高,达98.4%~100% ,与中国及东亚国家和地区的各个CGMMV 分离物cp 基因核苷酸序列相似 性也非常高,达98.6%~100% ,而与CGMMV 西班牙分离物和俄罗斯分离物cp 基因核苷酸序列相似性相对较低, 为 90.7%~92.6% 。从构建的系统关系树可以看出,17 个 CGMMV 安徽分离物与中国及东亚国家和地区的各个 CGMMV 分离物形成1 个单独的分支,而CGMMV 俄罗斯与西班牙分离物聚成另 1 个分支,说明来源于中国和东 亚的CGMMV 亲缘关系较近。 关键词:黄瓜绿斑驳病毒;cp 基因;克隆;序列分析 中图分类号:S436.421.1 文献标识码:A 文章编号:1672?352X (2016)04?0593?05 Molecular evolution analysis of cp genes of Cucumber green mottle mosaic virus (CGMMV) from different regions of Anhui Province LI Shuai, SHAN Wenshu, FENG Mingfeng, JIANG Xizi , JIANG Tong (School of Plant Protection, Anhui Agricultural University, Hefei 230036) Abstract: In order to reveal the molecular variation and phylogenetic evolution of Cucumber green mottle mosaic virus (CGMMV) isolated from Anhui Province, Cucurbitaceae plant samples infected with CGMMV were collected from five regions in Anhui province. Total RNA was extracted from the samples, and cp genes of 17 CGMMV isolates were obtained by RT-PCR amplification, cloning and sequencing.
