HPLC法测定合胃口服液中橙皮苷含量[摘要] 目的：建立合胃口服液中橙皮苷含量测定方法。方法：采用高效液相色谱法（HPLC）测定本品中橙皮苷的含量。ODS柱（250 mm×4.6 mm×5 μm），甲醇-2%乙酸水溶液（39:61）为流动相，流速为1 ml/min，检测波长为283 nm。结果：橙皮苷在0.134 4～1.680 0 μg含量呈线性关系，r=0.999 8；加样回收率为98.44%，RSD=1.21%（n=6）。结论：本法灵敏、准确、重现性好，可作为合胃口服液的质量控制指标之一。 [关键词] 橙皮苷；高效液相色谱法；合胃口服液；质量控制 [中图分类号] R286.0 [文献标识码] A [文章编号] 1674-4721（2012）01（a）-040-02 HPLC determination of content of hesperidin in combined stomach oral liquid ZHANG Ke Affiliated Hospital of Henan Institute of Traditional Chinese Medicine, Zhengzhou 450004， China [Abstract] Objective: To establish the determination method for determining the content of hesperidin in combined stomach oral liquid. Methods: The high-performance liquid chromatography (HPLC) method was used to determine the content of hesperidin in the studied product. The Octadecylsilyl (ODS) column was 250 mm×4.6 mm×5 μm; the methanol-2% acetic acid solution (39∶61) presented mobile phase; the flow rate was 1 ml/min; the detection wavelength was 283 nm. Results: Hesperidin showed a linear relationship in the content of 0.134 4 to 1.680 0 μg, r=0.999 8. The sample recovery rate was 98.44%, RSD=1.21% (n=6). Conclusion: This method is sensitive, accurate and reproducible, and therefore can be used as one of the quality control indicators of combined stomach oral liquid. [Key words] Hesperidin; HPLC; Combined stomach oral liquid; Quality control 合胃口服液是本院消化科常用制剂，由陈皮、藿香、砂仁、厚朴、鸡内金等组成，功能：理气合胃、消积导滞。陈皮为君药，具有行气、止呕、燥湿、化痰作用，橙皮苷为其主要有效成分[1]，笔者以《中国药典》2010版药典一部[2]陈皮项下橙皮苷的含量测定方法为基础，建立以橙皮苷成分的合胃口服液质量控制标准。本法可作为合胃口服液内在质量控制的定量指标。 1 仪器与试剂 Waters 2695高效液相色谱仪。Waters 2696二极管阵列检测器，Empower色谱工作站（美国Waters）。Hypersil ODS柱（250 mm×4.6 mm×5 μm） 橙皮苷（中国药品生物制品检验所，批号：0721-200010），甲醇（批号100821，天津四友精细化学品有限公司），水为重蒸馏水，其他所用试剂均为分析纯。合胃口服液由本院制剂室生产，生产批号依次为101103、101104、101105。 2 方法与结果 2.1 对照品溶液的制备 精密称取橙皮苷对照品8.40 mg，置25 ml容量瓶中加甲醇适量，超声溶解，加甲醇稀释至刻度，摇匀。精密吸取2 ml，置10 ml量瓶中，加甲醇2 ml混匀，加水稀释至刻度，摇匀，即得。每1毫升中含橙皮苷67.20 μg。 2.2 样品制备 2.2.1 供试样品制备 精密量取合胃口服液10 ml，加三氯甲烷提取
