蛋白酶的发酵工艺研究
摘要 中性蛋白酶是最早被发现并广泛应用于工业化生产的蛋白酶制剂,可用于皮革脱毛、软化、畜禽血液蛋白质水解、果酒、啤酒和饮料的澄清以及医学治疗等应用领域中。本文对一株产中性蛋白酶枯草芽孢杆菌(AS1.398)的发酵条件进行了研究,考察不同因素条件下对产酶的影响。通过摇瓶培养研究碳源浓度,不同接种量对蛋白酶产量的影响。利用酪蛋白培养基的平板实验观察蛋白酶的性质。利用发酵罐培养观察时间对产酶的影响.结果表明:豆饼粉3.0g/100ml,玉米粉4.0g/100ml,麸皮粉2.5g/100ml,磷酸氢二钠0.4g/100ml,磷酸二氢钾0.03g/100ml,pH 7.2-7.4为最适宜培养基组分。并且发酵罐培养的产酶活性高于摇瓶培养。

The Fermentation Process Research of Protease
Abstract Neutral protease is the first to be discovered and widely used in industrial production of the protease preparation ,which can be used for leather hair removal, softening, animal blood protein hydrolysis, wine, beer and beverage clarification, and in applications such as medical treatment.The fermentation conditions of a Bacillus Subtilis(AS1.398) which can produce neutral protease were studied in this paper. Under the condition of different factors to examine the influence of enzyme production.This study discussed concentration of carbon source, different inoculum on protease yield by shake flask. Casein petri dishes has been used in the observation of the nature of the protease. Fermentation tanks culture was used for observing times effection on the production of enzymes. the results showed that soybean meal 3.0g/100ml, corn flour 4.0g/100ml,wheat bran powder 2.5g/100ml,disodium hydrogen phosphate 0.4g/100ml,potassium dihydrogen phosphate 0.03g/100ml, pH 7.2-7.4 was the most appropriate medium composition. Enzyme production activity of Fermentation tanks culture was higher than shaking culture.

1.1 自然界中有大量产泌外蛋白酶微生物,其中以微生物的产酶活性最优。微生物来源蛋白酶一般按蛋白酶的最适pH值分为酸性、中性、碱性蛋白酶三类。中性蛋白酶最早被发现并广泛应用于工业化生产中。本实验采用枯草芽孢杆菌AS1.398做为研究菌株,其产酶量大,酶的耐热性好,因而被广泛应用于工业化生产中。此菌株在C/N为1:4左右时的产蛋白酶活性最高,并且有机N源优于无机N源,因而其发酵成本较高。探索新的发酵途径,寻找更加经济的组合方式显出了重大的意义。
1.2 此次实验主要做蛋白酶摇瓶发酵工艺的优化,改变发酵的C源浓度和接种量,测定蛋白酶酶活,以确定最优的发酵工艺条件。提取发酵液中的蛋白酶,测定酶活算出前后总活力,计算得
