Introduction:
Humanrespiratorysyncytialvirus(HRSV)isacommonpathogenthatcausesrespiratoryinfections,especiallyininfantsandyoungchildren.Currently,thereisnoeffectivevaccineorantiviraltherapyforHRSV.Therefore,thereisanurgentneedtodevelopasafeandeffectivevaccineagainstHRSV.
ThegoalofthisresearchprojectistoexplorepurificationconditionsforHRSVandevaluatetheimmunogenicityofexperimentalvaccines.TheprojectaimstoprovideimportantinsightsintothedevelopmentofanHRSVvaccine.
Methods:
Theresearchprojectwillbeconductedintwophases.Inthefirstphase,thepurificationconditionsofHRSVwillbeexplored.HRSVwillbepropagatedinHEp-2cells,andtheviruswillthenbepurifiedfromthecellculturesupernatantsbyultracentrifugation.ThepurityoftheviruswillbeassessedbySDSandWesternblotanalysis.
Inthesecondphase,experimentalvaccineswillbepreparedusingthepurifiedHRSV.Differentformulationsofexperimentalvaccineswillbetested,includinginactivatedvirusvaccines,subunitvaccines,andgeneticvaccines.Theimmunogenicityoftheexperimentalvaccineswillbeevaluatedinmicebymeasuringantibodytitersandcytokineresponses.
Expectedresults:
TheexplorationofpurificationconditionsforHRSVisexpectedtoresultintheidentificationofefficientandreliablemethodsforviruspurification.Thiswillenabletheproductionofhigh-qualityexperimentalvaccinesforfurthertesting.
ThepreliminaryevaluationoftheimmunogenicityofexperimentalvaccinesisexpectedtoprovideimportantinsightsintothedevelopmentofanHRSVvaccine.TheresultsobtainedfromthisstudywillfacilitatethedesignofmoreeffectiveandsafeHRSVvaccinesinthefuture.
