高血压大鼠肾脏中基质金属蛋白酶及组织型基质金属蛋白酶抑制剂表达与活性的变化 姚旻1 曾山2 涂悦3 庞伟2 朱勇4李红涛1 (1.武警天津总队医院内科,天津,300162;2.武警后勤学院附属医院心血管病研究所,天津,300162;3.武警后勤学院附属医院脑系科中心,天津,300162;4.武警湖北总队医院心内科,武汉,430061) 中文摘要 研究在卒中易感自发性高血压大鼠(Stroke-Prone Spontaneously Hypertensive Rat, SHR-SP)中基质金属蛋白酶(Matrix metalloproteinases,MMPs)-2,-9以及组织型基质金属蛋白酶抑制剂(Tissue Inhibitors of Matrix Metalloproteinases, TIMPs)-1,-2活性的。方法:0只7周龄雄性SHR-SP大鼠同周龄雄性Wistar-Kyoto大鼠阴性对照。观察期间。明胶酶谱法分析存活大鼠MMP-2和MMP-9活性采用逆明胶酶谱法及蛋白质印迹分析TIMP1和TIMP-2表达和活性结果:(1)明胶酶谱分析显示WKY组明显增高(3.281.17 vs. 1.260.62; p0.01)。(2)明胶酶谱显示:高血压大鼠TIMP-2的活性明显高于WKY组(0.43 vs. 0.780.34, p0.05)。()蛋白质印迹结果显示:组TIMP-1、TIMP-2表达较对照组(3.44±0.13 vs. 2.31±0.15 ,TIMP-2:6.44±0.49 vs. 4.94±0.68,p 0.01)结论: SHR-SP组织中MMP-2。关键词:基质金属蛋白酶;基质金属蛋白酶抑制剂R363.2 The Expression and the Activity of Renal Matrix Metalloproteinases and Tissue Inhibitors of Matrix Metalloproteinases in the Stroke-prone Spontaneously Hypertensive Rats Yao Min 1 Zeng Shan2 Tu Yue 3 Pang Wei2 Zhu Yong4 Li Hongtao1 (1.Department of Internal Medicine, Tianjin Corps Hospital of Chinese People’s Armed Police Forces,Tianjin 300162 China. 2. Abstract Abjective The purpose of this research was to investigate the expression and the activity of renal matrix metalloproteinases and the tissue inhibitors of matrix metalloproteinases in stroke-prone spontaneously hypertensive rats (SHR-SPs).Methods 40 male SHR-SPs of 7 weeks old were vehicle group and 10 male Wistar-Kyoto rats (WKY) at the same age were used as controls.
