转化生长因子β1对钛表面成骨细胞因子表达的影响
【摘要】目的:通过观测特定浓度的转化生长因子β1(transforming growth factors β1,TGF-β1)对钛表面成骨细胞中骨钙素(osteocalcin,OC)、骨涎蛋白(bone sialoprotein,BSP)和I型胶原蛋白(collagen-I,COL-I)表达的影响,以探讨其对钛表面成骨细胞增殖分化的作用。方法:体外培养成骨细胞接种于钛片表面,将其分为刺激组和对照组,刺激组加入0.5ng/ml的TGF-β1刺激细胞增殖和分化,收集第1d、3d、7d培养的细胞。通过RT-PCR检测不同培养时间OCmRNA、BSPmRNA和Col-ImRNA表达情况,对照组不加TGF-β1。实验中所有数据采用SPSS 10.0软件作统计学处理分析,比较各组间差异程度,P 0.05认为差异有显著性意义。结果:浓度为0.5ng/ml的外源性TGF-β1加入到接种于钛片表面的成骨细胞中后,均可显著增加几种细胞因子(OC、BSP和Col-I)的表达(P0.05)。结论:浓度为0.5ng/ml的外源性TGF-β1对钛片表面的成骨细胞成骨相关因子表达有一定的促进作用。

The effects of TGF-β1 on the gene expression of osteoblasts on Titanium disc surfaces in vitro

[Abstract]Objective: The purpose of this study is to investigate the consequential changes of gene expression of osteoblasts on Titanium disc surfaces incubated with TGF-β1,and to make a primary understanding of functions of proliferation and differentiation.Methods: Osteoblast were divided into two groups: the control group and the induced group. The cells were seeded on surfaces of Ti disc and composed of Dexamethasone(DEX), β-phosphoglycerol and Vitamin C. TGF-β1 with a dose of 0.5ng/ml was administrated to induced group.The expression of mRNA were detected by reverse transcription-polymerase chain reaction(RT-PCR).All the data were analyzed by SPSS10.0 software.Results:The extraneous TGF-β1 (0.5ng/ml) can increase the gene expressions(OC,BSP and Col-I) of osteoblasts on Ti disc surfaces(P0.05).Conclusions:It is concluded that extraneous TGF-β1 with a dose of 0.5ng/ml increased osteoblase gene expressions on the surfaces of Ti disc.
