碱性成纤维细胞生长因子对牙周膜细胞表皮生长因子受体基因表达的影响 碱性成纤维细胞生长因子对牙周膜细胞表皮生长因子受体基因表达的影响 作者:陈莹 林崇韬 高颖 作者单位:吉林大学口腔医院 牙周病科,吉林 长春 130041 【摘要】 目的 研究碱性成纤维细胞生长因子(bFGF)对人牙周膜细胞(PDLC)表达表皮生长因子受体(EGFR)的影响,探讨bFGF在牙周组织分化再生中的意义。方法 体外原代培养人PDLC,有限稀释法形成单细胞克隆,用外源性bFGF刺激单细胞克隆,采用逆转录聚合酶链反应(RT-PCR)检测克隆细胞内EGFR基因表达的变化。结果 bFGF促进人PDLC内EGFR mRNA的合成,并且随着质量浓度的增加促进作用增强。结论 bFGF对EGFR的促进作用很可能是牙周炎损伤修复过程中一个重要的调节因素,为牙周组织分化再生提供部分理论基础。 【关键词】 碱性成纤维细胞生长因子 表皮生长因子受体 牙周膜细胞 细胞分化   Effect of basic fibroblast growth factor on the gene expression of epidermal growth factor receptor by pe-riodontal ligament cells in culture CHEN Ying, LIN Chong-tao, GAO Ying. (Dept. of Periodontology, StomatologyHospital of Jilin University, Changchun 130041, China)   [Abstract] Objective To study the effect of basic fibroblast growth factor(bFGF) on the gene expression of epi-dermal growth factor receptor(EGFR) by periodontal ligament cells(PDLC) in culture and discuss the effect of bFGF in cell differentiation and periodontal regeneration. Methods Human PDLC were cultured in vitro and cell clone was obtained by the method of limiting dilution. PDLC were stimulated by bFGF, and then gene expression of EGFR was assessed by reverse transcription-polymerase chain reaction(RT-PCR). Results The mRNA expression of EGFR waspromoted by bFGF and the effect was dose-dependent. Conclusion The promotion effect of the mRNA expression of EGFR in PDLC may be one of important factors which participate in the healing process of periodontitis and provide partly theoretical basis of cell differentiation and periodontal regeneration.   [Key words] basic fibroblast growth factor; epidermal growth factor receptor; periodontal ligament cells; cell differentiation   牙周膜细胞(periodontal ligament cells,PDLC)的表型分化对牙周组织在2种硬组织之间维持自身稳定性起重要作用。在牙周组织再生过程中,牙周膜细胞的表型分化决定治疗的预后和转归。碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)是肝素黏合多肽,分布于多种组织中,它是重要的有丝分裂促进因子,也是形态发生和分化的诱导因子[1]。表皮生长因子受体(epidermal growthfactor receptor,EGFR)是牙周膜细胞成纤维表型的标记物[2]
