·874 · 中草药 Chinese Traditional and Herbal Drugs 第49 卷 第4 期 2018 年2 月 黄连水提物对秀丽隐杆线虫毒性效应评价 * * 苗祥贞，张 潇，袁炎炎，张亚丽，高 简，刘永刚 ，谭 鹏 北京中医药大学，北京 102488 摘 要：目的 评价不同质量浓度黄连水提物对秀丽隐杆线虫的毒性效应。方法 制备黄连水提物，以秀丽隐杆线虫为模式 生物，通过测定不同质量浓度的黄连水提物对秀丽隐杆线虫死亡率、最长寿命与半数致死时间、个体发育、产卵数和运动行 为的影响，评价黄连水提物对秀丽隐杆线虫的毒性效应。结果 与对照组相比，黄连水提物0.5、1.0、2.5 mg/mL 可使秀丽 隐杆线虫的死亡率显著提高 （P ＜0.01 ），最长寿命与半数致死时间显著降低（P ＜0.01 ）；在个体发育方面，黄连水提物1.0、 2.5 mg/mL 能显著抑制线虫生长（P ＜0.01 ）；生殖行为方面，3 个质量浓度的黄连水提物均能显著降低秀丽隐杆线虫产卵数 （P ＜0.001 ），且具有一定的浓度依赖性；在运动行为方面，3 个质量浓度的黄连水提物均能显著降低秀丽隐杆线虫头部摆动 频率（P ＜0.01 ），黄连水提物0.5 、1.0 mg/mL 组线虫身体弯曲频率无显著性差异，黄连水提物2.5 mg/mL 组线虫身体弯 曲频率极显著降低（P ＜0.01 ）；3 个质量浓度的黄连水提物组线虫向前摆动、向后摆动和 Omega/U 摆动频率均无显著性 差异。结论 黄连水提物对秀丽隐杆线虫具有明显的毒性作用，为以秀丽隐杆线虫为模式生物评价黄连不同炮制品毒性提 供了依据，为中药毒性的生物评价提供了新的思路和方法。 关键词：黄连；秀丽隐杆线虫；死亡率；寿命；运动行为；毒性评价 中图分类号：R285.5 文献标志码：A 文章编号：0253 - 2670(2018)04 - 0874 - 05 DOI: 10.7501/j.issn.0253-2670.2018.04.020 Toxicity evaluation of water extracts of Coptidis Rhizoma on Caenorhabditis elegans MIAO Xiang-zhen, ZHANG Xiao, YUAN Yan-yan, ZHANG Ya-li, GAO Jian, LIU Yong-gang, TAN Peng Beijing University of Chinese Medicine, Beijing 102488, China Abstract: Objective To evaluate the toxic effects of different concentrations of Coptidis Rhizoma extract on Caenorhabditis elegans.
