摘要:
脑胶质瘤是一种常见的致死性肿瘤,其高度浸润性和恶性生长方式
使得治疗非常困难。因此,研究其发病机制以及寻找治疗靶点显得尤为
重要。本文选取了GOLPH3作为研究对象,通过实验探究了GOLPH3对
脑胶质瘤细胞迁移的作用及机制。
首先,实验结果表明在人脑胶质瘤组织中,GOLPH3的表达水平比
正常脑组织明显升高。进一步地,通过使用GOLPH3-siRNA干扰法来减
少GOLPH3在脑胶质瘤细胞中的表达水平,发现GOLPH3-siRNA组脑胶
质瘤细胞的迁移能力较对照组明显降低。此外,实验结果还显示减少
GOLPH3表达可降低脑胶质瘤细胞中细胞骨架蛋白F-actin和α-actinin的
表达水平。
综上,实验表明GOLPH3在脑胶质瘤细胞中发挥作用,可以促进其
迁移能力并影响其细胞骨架的表达水平。研究结果为进一步探究脑胶质
瘤的发病机制,以及开发新的治疗靶点提供了新的思路和方向。

Abstract:
Glioblastomaisacommonfataltumor,anditshighlyinfiltrative
andmalignantgrowthmodemakestreatmentverydifficult.Therefore,
studyingitspathogenesisandfindingtreatmenttargetsisparticularly
important.Inthispaper,GOLPH3wasselectedastheresearchobject,
andtheeffectandmechanismofGOLPH3onthemigrationofglioma
cellswereexploredthroughexperiments.
Firstly,theexperimentalresultsshowedthattheexpressionlevel
ofGOLPH3inhumanglioblastomatissuewassignificantlyhigherthan
thatinnormalbraintissue.Furthermore,byusingGOLPH3-siRNA
interferencemethodtoreducetheexpressionlevelofGOLPH3in
glioblastomacells,itwasfoundthatthemigrationabilityof
glioblastomacellsintheGOLPH3-siRNAgroupwassignificantlyreduced
comparedwiththecontrolgroup.Inaddition,theresultsshowedthat
reducingtheexpressionofGOLPH3canreducetheexpressionlevelsof
F-actinandα-actinin,whicharecellskeletonproteins,inglioblastoma
cells.
Inconclusion,theexperimentshowedthatGOLPH3playsarolein
glioblastomacells,canpromoteitsmigrationability,andaffectthe
expressionlevelofitscellskeleton.Theresearchresultsprovidenew
ideasanddirectionsforfurtherexploringthepathogenesisof
glioblastomaanddevelopingnewtreatmenttargets.
