摘要:目的:探讨 NRP1 与不同种类非小细胞肺癌细胞株放射敏感性的关系,并筛选辐射抗
性最强的细胞株,以便进一步研究 NRP1 在非小细胞肺癌产生辐射抗性中的作用机制。方法:
通过 Western blot 技术检测不同来源的 5 种人非小细胞肺癌细胞株(H358、H460、H1299、
A549、SK-MES-1) NRP1 的基础表达水平,使用深部 X 光机对 5 株细胞进行不同剂量照射,
通过 MTT 实验比较肿瘤细胞的辐射敏感性,初步筛选辐射敏感性最高与最低的 2 种细胞;
进一步采用克隆形成实验、划痕实验及流式细胞术分别检测细胞存活分数、迁移能力及凋亡
率的变化,分析 2 种细胞克隆形成能力、迁移及凋亡率与细胞放射敏感性的关系,并采用
Western blot 检测照射后 2 种细胞 NRP1 的表达变化。结果:5 种肿瘤细胞 NRP1 表达量
由高到低依次为:A549 SK-MES-1 H358 H460H1299;X 射线对肿瘤细胞的抑制能力随
着照射剂量的增加而增强,其抑制率由强到弱依次为:H460 H1299 H358 SK-MES-1
A549,因此选取 A549 细胞和 H460 细胞做后续研究。克隆形成实验显示,A549 在 2 Gy
照射下的存活分数(SF2)是 0.8872,而 H460 细胞为 0.3134。X 射线对 A549 的迁移抑制
力弱于 H460,10 Gy X 射线作用后 H460 细胞的凋亡率明显高于 A549 细胞,并且受照后
A549 细胞 NRP1 的表达量上调。结论:在非小细胞肺癌细胞中 A549 的辐射敏感性最弱,
且辐射抗性的形成可能与 NRP1 表达上调有关。

Abstract: Objective: To explore the relation of NRP1with the radiosensitivity of different types of
non-small cell lung cancer. Methods: The expression of NRP1on the cells of H358、H460、H1299、
A549 and SK-MES-1was detected by Western blot .MTT was used to detect cell proliferation, and
selected two cells which were the highest and lowest radiosensitivity ;The SF、Migration and
apoptosis rate was detected with Clonogenic assay, wound healing assay and flow cytometry
respectively. Results:The expression levels of NRP1 in the five cell lines (A549 SK-MES-1
H358 H460 H1299) were all increased by degrees in order.The radiosensitivity of five NSCLC
cell lines were showed a dose-dependent decrease (A549SK-MES-1H358 H1299
H460).According to the levels of NRP1and the radiosensitivity, we selected A549 cells and H460
cells to study deeply. The values of survival fraction at 2 Gy (SF2)were 0.8872 in A549 cells,
