非小细胞肺癌中基质金属蛋白酶10的表达 作者:朱少伟,罗光华,张晓膺 作者单位：苏州大学附属第三医院1.心胸外科； 2.综合实验室，江苏 常州 213003 【摘要】 目的： 探讨非小细胞肺癌(nonsmall cell lung cancer, NSCLC)中基质金属蛋白酶10(matrix metalloproteinase10,MMP10）的表达及其与临床参数间的关系。方法： 使用RTPCR法及免疫组化法，检测32对NSCLC癌组织及癌旁正常组织中MMP10的表达水平,并分析其与性别、年龄、分期、分级、肿瘤大小、淋巴结转移及病理类型之间的相关性。结果： MMP10 的表达在mRNA水平与蛋白质水平表达有差异： MMP10 mRNA在NSCLC癌组织中的表达要显著低于癌旁正常肺组织（P0.05），MMP10蛋白在肺癌癌组织中的表达要显著高于癌旁正常肺组织（P0.01）；在NSCLC癌组织中MMP10 mRNA 的表达与MMP10蛋白的表达呈正相关(r=0.4672, P0.05),而在癌旁正常肺组织中,两者表达相关性无显著统计学差异(r=-0.003022,P0.05)。结论： MMP10 mRNA在肺癌癌组织中表达要低于正常组织，MMP10蛋白在肺癌癌组织中表达要高于正常组织。 【关键词】 非小细胞肺癌; 基质金属蛋白酶10; PCR; 免疫组化 　　Expression of MMP10 in nonsmall cell lung cancer 　　ZHU ShaoWei1, LUO GuangHua2, ZHANG XiaoYing1, ZHENG Lu2, WEI Jiang2, ZHU Jiang2 　　 　　(1.Department of Thoracic and Cardiovascular Surgery, 2.Laboratory of Molecular Medicine, the Third Affiliated Hospital, Suzhou University, Changzhou Jiangsu 213003, China) 　　［Abstract］ Objective: To analyze matrix metalloproteinase10 (MMP10) expression in nonsmall cell lung cancer (NSCLC), evaluate the correlations between MMP10 expression and clinicopathologic parameters. Methods： RTPCR was used to assess MMP10 mRNA expression while immunohistochemistry was used to assess MMP10 protein expression in 32 matched lung tumor tissues and corresponding adjacent normal lung tissues. The correlations between MMP10 mRNA, protein expression and clinical parameters, such as gender, age, stage, tumor size, lymph node metastasis or histological type was analyzed. Results： There was difference between MMP10 mRNA expression and MMP10 protein expression in NSCLC. MMP10 mRNA level was lower in tumor tissue than that in corresponding normal lung tissue by Realtime RTPCR (P0.05).
