泥蚶谷胱甘肽过氧化物酶基因的克隆和分析
摘 要
本实验通过RACE法获得泥蚶谷胱甘肽过氧化物酶(glutathione peroxidase)基因cDNA全长序列并对其进行生物信息学分析。发现,泥蚶谷胱甘肽过氧化物酶不含硒,称为过氧化物酶6(TgPrx6)。泥蚶过氧化物酶6基因的全长cDNA为1937个碱基,其中开放阅读框为672个碱基,编码224个氨基酸。其次,用RNAfast200总RNA极速抽提试剂盒从泥蚶血液和其它组织中提取RNA,逆转录成cDNA。最后,采用PCR方法分析TgPrx6基因在不同组织中的表达情况,结果表明TgPrx6基因的组织表达有所差异。TgPrx6基因在内脏团中表达水平最高,其次是鳃和外套膜,在血液中的表达水平最低。

ABSTRACT
A whole length cDNA of glutathione peroxidase in Tegillarca granosa was got by RACE and the sequenced of bioinformatics was analysed. The glutathione peroxidase of Tegillarca granosa did not contain Se, which was called peroxidase 6(TgPrx6).The whole length cDNA of TgPrx6 in Tegillarca granosa was 1937 bp, and the open reading frame was 672bp which could translate 224 amino acids. RNA was got by RNAfast200 Kit from blood and other tissues of Tegillarca granosa, then RNA was reversed transcriptase to cDNA. Finally, the gene of TgPrx6 was analysed to explore how the gene of TgPrx6 expressed in different tissues, and the result was that there were differences between tissues. The highest level of TgPrx6 expression was detected in viscera, and then gills and mantle. The lowest level was detected in blood .
