转录因子CEBPB激活FJX1促进结肠癌细胞的增殖、侵袭、迁移和血管生成能力的研究

[摘要]?目的?探究CCAAT增强子结合蛋白B(CCAAT?enhancer?binding?protein?beta,CEBPB)与四连接同源基因(four-jointed?box?kinase?1,FJX1)在结肠癌(colon?cancer,CC)中的调控关系及其对CC恶性进展及血管生成的影响。方法?通过生物信息学分析FJX1和CEBPB在CC中的表达情况及二者之间的调控关系。利用实时荧光定量核酸扩增检测系统(real-time?quantitative?reverse?transcription?polymerase?chain?reaction,qRT-PCR)验证FJX1与CEBPB在CC细胞中的表达情况,利用染色质免疫沉淀(chromatin?immunoprecipitation,CHIP)与双荧光素酶实验验证FJX1与CEBPB之间的结合关系。利用細胞计数试剂-8(cell?counting?kit-8,CCK-8)、划痕试验、Transwell和血管形成实验检测FJX1与CEBPB对CC细胞增殖、迁移、侵袭及血管形成能力的影响。结果?本研究发现FJX1在CC中高表达,抑制FJX1的表达会显著抑制结肠癌细胞的细胞增殖、迁移、侵袭及血管形成能力。CEBPB是FJX1的上游调控基因,且CEBPB在结肠癌中高表达。CHIP与双荧光素酶实验结果显示,CEBPB可与FJX1相结合。细胞实验结果表明,转录因子CEBPB可通过激活FJX1的表达,从而促进CC细胞的增殖、迁移、侵袭及血管生成。结论?CEBPB/FJX1轴在CC的进展中发挥促癌作用,提示CEBPB和FJX1可能是CC的潜在治疗靶点。
