摘要:
【背景】热疗是通过加热使肿瘤组织的温度达到40~44℃,引起肿瘤细胞生长受阻与死亡的一种治疗方式,热疗对细胞的杀伤作用取决于热疗的温度及热疗的时间。联合应用热疗与化疗是一种合理的综合治疗方案,热疗能促进药物进入肿瘤细胞;促进药物诱发的肿瘤细胞凋亡;肿瘤组织中心部位达到较高温度时的酸性环境易诱发肿瘤细胞凋亡。
【目的】本实验旨在国内外现有的研究基础上,探讨数种抗肿瘤药物联合热疗体外抑制人肝癌SMMC-7721细胞???殖的效果,以确定联合方案是否具有协同效应。
【方法】本实验使用MTT法测定单独或联合应用化疗药物羟基喜树碱或顺铂与热疗对人肝癌SMMC-7721细胞的体外增殖的抑制作用,使用流式细胞仪检测了不同方案对SMMC-7721细胞凋亡率的影响。
【结果】结果表明,联合应用热疗可以显著提高化疗药物顺铂或羟基喜树碱引起的细胞增殖抑制及细胞凋亡。以亚毒性剂量浓度的顺铂为例,对照组、化疗组、热疗组、热化疗组的细胞增殖抑制率分别为0%、20.77%、32.46%、62.76%;细胞倍增时间分别为40.54h、50.30h、106.85h、166.07h;细胞凋亡率分别为2.19%、10.56%、15.16%、34.32%。
【结论】联合应用热疗与化疗药物羟基喜树碱或顺铂,对SMMC-7721细胞体外增殖的抑制及诱导细胞凋亡的能力具有显著的协同作用。

Abstract:
【Background】Hyperthermia is an attractive addition to multidisciplinary approaches to clinical cancer treatment. The quantitative and reproducible effect of hyperthermia depends on the elevation of temperature and the duration of treatment. It has been reported that in vitro and in vivo hyperthermia can enhance the cytotoxic effect of certain anticancer drugs that have different mechanisms of action. Hyperthermia combined with chemotherapy is a logical and potentially powerful treatment. The magnitude of the enhancement varies according to different drugs and the scheduling of drugs with heat.
【Objectives】This experiment aims to investigate whether concurrent exposure of human hepatocellular carcinoma cells SMMC-7721 to chemotherapeutic agents with hyperthermia will increase anticancer effects.
【Methods】In our study, we applied two chemotherapeutic agents such as cisplatin, and hydroxycamptothecin. The MTT assay has been developed for quantitative evaluation of the proliferation of SMMC-7721 and cytotoxicity of anticancer drug with or without hyperthermia. Flow cytomet
