胡桃醌体内抗肿瘤作用研究

摘 要 目的 研究胡桃醌的体内抗肿瘤作用。方法 建立S180肉瘤模型,考察胡桃醌对S180荷瘤小鼠的抑瘤率和胸腺指数、脾脏指数的影响;HE染色,观察S180肉瘤的病理改变;建立H22肝癌腹水瘤模型,考察胡桃醌对H22荷瘤小鼠生命延长率的影响。结果 在0.25 mg?kg-1·d-1、0.5 mg?kg-1·d-1、1 mg?kg-1·d-1剂量下,胡桃醌对S180实体瘤的抑瘤率分别为17.09%、22.12%、29.61%,尤其是0.5 mg?kg-1?d-1剂量和1.0 mg?kg-1?d-1剂量抑瘤作用明显(P0.01);0.5 mg?kg-1?d-1、1.0 mg?kg-1?d-1剂量的胡桃醌使脾脏指数降低(P0.05,P0.01),各剂量的胡桃醌胸腺指数无明显变化(P0.05);光镜下胡桃醌各组肿瘤组织坏死程度明显增加。在0.25 mg?kg-1·d-1、0.5 mg?kg-1·d-1、1 mg?kg-1·d-1剂量下,胡桃醌对H22荷瘤小鼠的生命延长率分别为73.88%、82.09%、99.25%,与空白对照组比较有显著性差异(P0.05)。结论 胡桃醌体内有明显的抗肿瘤作用,能有效抑制肿瘤生长,延长荷瘤小鼠生命。

ABSTRACT Objective In this experiment, the anti-tumor effects of Juglone was studied on tumor mice. Methods The graft mouse tumor model S180 was used to investigate the effects of juglone on tumor growth and the weights of immune organs. After HE staining, the growth and patho-morphologic changes of tumor cells were observed by optical microscope. The graft mouse tumor model H22 was used to investigate the effects of juglone on survival time. Results The inhibitory rate of juglone at the dosage of 0.25, 0.5 and 1 mg?kg-1·d-1 was 17.09%, 22.12%, 29.61% respectively. The inhibitory rate of juglone at the dosage of 0.5, 1 mg?kg-1·d-1 was significantly increased compared with the control group (P0.01). Spleen index in 0.5, 1 mg?kg-1·d-1 dosage group had significant decrease (P0.05, P0.01), and Thymus index had no change.
