摘 要
蛹虫草,也称北冬虫夏草,具有补虚损、益精气、保肺、益肾、止血化痰、滋补强壮等功效,是我国的名贵中草药材。
本论文采用沈阳微生物研究所提供的蛹虫草优质菌种,通过斜面培养、液体培养等过程,获得优质的蛹虫草菌丝体,并以菌丝体重量为指标,对培养温度、初始pH值、培养箱转速、最佳碳源、最佳氮源、最佳无机盐等液体培养条件进行单因素分析,通过正交试验法确立了蛹虫草菌丝体的最佳培养基配方。实验结果表明,蛹虫草菌丝体培养最佳碳源是葡萄糖,最佳氮源是蛋白胨,最佳无机盐是KH2PO4和MgSO4;最佳培养基配方是葡萄糖20g/L,蛋白胨10g/L,KH2PO4 1.0g/L,MgSO4 0.5g/L,VB1 50mg/L;最佳培养温度是24℃,最佳初始pH值是6.0,最佳培养箱转速时150r/min;并在最佳培养条件下绘制了蛹虫草菌丝体生长曲线,其对数期为第5~7天,菌丝体平均重为7.07g,稳定期为第7~9天,菌丝体重为14.63g,从第9天后,蛹虫草菌丝体开始衰退。
Abstract
Cordyceps militaris , also known as the North Cordyceps, has the effect of completing deficiency, benefiting the essence, lung and kidney, haemostatic and resolving phlegm, nourishing and strong, It is a precious Chinese herbal medicine materials.
Cordyceps militaris in this paper has high quality and is provided by the Shenyang Institute of Microbiology. After taking slant culture and liquid culture processes, we get high quality Cordyceps militaris mycelia. It takes mycelium weight as an indicator to make univariate analysis of the incubation temperature, initial pH value incubator speed, the best carbon source, the best nitrogen source and the best inorganic salts liquid culture conditions. With orthogonal test method, it’s established the optimum mycelia medium. The experimental results show that the best carbon source for the mycelia is glucose, the best nitrogen source is peptone, the best inorganic salt is KH2PO4 and MgSO4.A optimum medium includes glucose 20g/L, peptone 10g/L, KH2PO4 1.0g/L, MgSO40.5g/L,VB1 50mg/L.The optimal incubation temperature is 24℃, the optimal initial pH value is 6.0, the best incubator speed is 150r/min. The growth curve of Cordyceps militaris mycelia is drawn on the optimal culture conditions. The logarithmic phase turned up at the 5th to 7.5th day, the mycelium average weighted 7.07g, the stabilization period turned up at the 7.5th to 9th day. The mycelium weighted 14.63g and the recession began from the ninth day.
