摘要：

亚型兔出血症病毒（RHDV）是主要感染野兔的致命性病毒，其在国内野兔、家兔中被广泛发现。为了控制RHDV的传播和感染，需要建立有效的诊断和预防手段。本研究旨在制备不同亚型RHDV的单克隆抗体，并鉴定其抗原表位，为RHDV检测和疫苗研发提供理论依据。

首先采用RHDVCp抗原免疫BALB/c小鼠，制备单克隆抗体。通过ELISA筛选，获得5株高效、特异的单克隆抗体。进一步通过Westernblotting和间接免疫荧光法，证实了这些抗体对不同亚型RHDV的高效识别。随后，通过Peptidearray分析和毒株差异性研究，初步鉴定了抗原表位。结果表明，这些抗体的抗原表位主要位于RHDVCp蛋白的C端。

本研究制备了高效、特异的不同亚型RHDV单克隆抗体，并初步鉴定了其抗原表位。这些结果为RHDV检测和疫苗研发提供了理论依据，也为其他病毒的单克隆抗体制备和抗原表位分析提供了参考。

Abstract：

Rabbithemorrhagicdiseasevirus(RHDV)isafatalvirusthatinfectswildrabbitsandiswidelyfoundindomesticrabbitsinChina.InordertocontrolthespreadandinfectionofRHDV,itisnecessarytoestablisheffectivediagnosticandpreventivemeasures.Thisstudyaimstopreparemonoclonalantibodies(mAbs)againstdifferentsubtypesofRHDVandidentifytheirepitopestoprovidetheoreticalbasisforRHDVdetectionandvaccinedevelopment.

Firstly,BALB/cmicewereimmunizedwithRHDVCpantigentopreparemAbs.ThroughELISAscreening,5high-efficiencyandspecificmAbswereobtained.FurtherWesternblottingandindirectimmunofluorescenceassaysdemonstratedthatthesemAbscouldefficientlyrecognizedifferentsubtypesofRHDV.Subsequently,throughpeptidearrayanalysisandstraindifferentiationstudies,theepitopeswerepreliminarilyidentified.TheresultsshowedthattheepitopesofthesemAbsweremainlylocatedattheC-terminusofRHDVCpprotein.

ThisstudypreparedefficientandspecificmonoclonalantibodiesagainstdifferentsubtypesofRHDVandpreliminarilyidentifiedtheirepitopes.TheseresultsprovidetheoreticalbasisforRHDVdetectionandvaccinedevelopment,andalsoprovidereferenceforthepreparationofmonoclonalantibodiesandepitopeanalysisofotherviruses.
