Introduction:
Prostatecancerisoneofthemostcommonmalignanciesinmenworldwide,withitsincidenceandmortalityrateincreasingyearbyyear.Metastasis,invasionandangiogenesisaremajorfeaturesofadvancedprostatecancer,whichcontributetoitspoorprognosis.TheRUNX3gene,encodingatranscriptionfactor,hasbeenidentifiedasanoveltumorsuppressorgeneinvarioushumancancers.PreviousstudieshaveindicatedthatRUNX3geneexpressionisdecreasedinprostatecancertissues,anditsdownregulationisassociatedwithtumorprogressionandmetastasis.However,thepreciserolesandmolecularmechanismsofRUNX3geneinprostatecancercellmigration,invasionandangiogenesisremainlargelyunknown.
Objectives:
1.ToinvestigatetheexpressionlevelsofRUNX3geneinprostatecancertissuesanditscorrelationwithclinicopathologicalparameters.
2.ToexploretheeffectsofRUNX3geneoverexpressionorknockdownonprostatecancercellmigration,invasionandangiogenesisinvitro.
3.ToelucidatetheunderlyingmolecularmechanismsofRUNX3gene-mediatedregulationofprostatecancerprogression.
Methods:
1.QuantitativeRT-PCRandWesternblottingwillbeusedtodetecttheexpressionlevelsofRUNX3geneinprostatecancertissuesandcelllines.
2.CellmigrationandinvasionassayswillbeperformedtoassesstheeffectofRUNX3geneoverexpressionorknockdownonprostatecancercellmotilityandinvasiveness.
3.TubeformationassaywillbeusedtoevaluatetheeffectofRUNX3geneonangiogenesis.
4.Immunofluorescencestainingandco-immunoprecipitationwillbeperformedtoinvestigatethepotentialmolecularmechanismsofRUNX3geneinregulatingprostatecancerprogression.
