摘要
乙醇是一种常见的毒性物质,长期过量饮酒会导致肝脏氧化损伤。本研究旨在探究microRNA在乙醇诱导肝细胞氧化损伤和羊水干细胞肝向分化中的作用。河南省某医院的均龄为30岁的志愿者30名,半数男性和半数女性,饮酒量都在每日饮酒标准以下。首先,通过体外实验模拟肝细胞氧化损伤情况,比较乙醇处理前后肝细胞microRNA表达情况。其次,采用羊水干细胞的外泌体对诱导肝细胞进行治疗,观察外泌体中microRNA的表达情况及治疗效果。最后,通过实验验证特定microRNA是否能够影响羊水干细胞的肝向分化。

Abstract
Ethanolisacommontoxicsubstance,andlong-termexcessivedrinkingcancauseoxidativedamagetotheliver.ThisstudyaimstoexploretheroleofmicroRNAinethanol-inducedlivercelloxidativedamageandamnioticfluidstemcellhepaticdifferentiation.30volunteerswithanaverageageof30,halfmaleandhalffemale,andadrinkingamountbelowthedailydrinkingstandardwererecruitedfromahospitalinHenanProvince,China.First,aninvitroexperimentwasusedtosimulatelivercelloxidativedamage,andtheexpressionofmicroRNAwascomparedbeforeandafterethanoltreatment.Secondly,theextracellularvesiclesfromamnioticfluidstemcellswereusedforthetreatmentofinducedlivercells,andtheexpressionofmicroRNAintheextracellularvesiclesandthetreatmenteffectwereobserved.Finally,theexperimentwasusedtoverifywhetherspecificmicroRNAscanaffectamnioticfluidstemcellhepaticdifferentiation.
ThisstudyaimstoexploretheroleofmicroRNAinethanol-inducedlivercelloxidativedamageandamnioticfluidstemcellhepaticdifferentiation,providingnewtheoreticalbasisandexperimentalbasisforethanol-relatedliverinjuryandstemcelltherapy.
