Background:
Alcoholconsumptionhasbeenlinkedwithvariousphysiologicalandpsychologicalconditions,includingdecreasedbonedensityandincreasedriskoffractures.Thebonemarrowisaprimarytissueaffectedbyalcoholconsumption,asitisresponsibleforhematopoiesisandcangiverisetomesenchymalstemcells(MSCs).MSCshavetheabilitytodifferentiateintovariouscelltypes,includingosteoblasts,whichareresponsibleforboneformation.TheimpactofethanolonthefunctionsofMSCsandosteoblastsisstillnotfullyunderstood.
Objective:
Thisstudyaimstoinvestigatetheeffectsofethanolontheexpressionofneuropeptidesandapoptosis-relatedgenesinhumanbonemarrowmesenchymalstemcells.
Methods:
Humanbonemarrowmesenchymalstemcellswillbeisolatedandculturedinvitro.Thecellswillbetreatedwithdifferentconcentrationsofethanolfordifferenttimeperiodstomimicchronicalcoholexposure.Theexpressionofneuropeptides,includingsubstanceP,calcitoningene-relatedpeptide(CGRP),andtyrosinehydroxylase(TH),willbemeasuredusingquantitativepolymerasechainreaction(qPCR)andenzyme-linkedimmunosorbentassay(ELISA).Apoptosis-relatedgenes,includingBcl-2,Bax,andcaspase-3,willalsobemeasuredusingqPCR.
ExpectedOutcomes:
Itisexpectedthatethanolexposurewillleadtochangesintheexpressionofneuropeptidesandapoptosis-relatedgenesinhumanbonemarrowmesenchymalstemcells.Specifically,itishypothesizedthatethanolexposurewilldecreasetheexpressionofneuropeptidesandincreasetheexpressionofapoptosis-relatedgenes,whichcouldultimatelyleadtodecreasedboneformationandincreasedriskofbonefractures.
Conclusion:
Theresultsofthisstudywillprovideimportantinsightsintotheimpactofethanolonthefunction
