SFRP2基因表观遗传沉默在食管鳞癌中的作用机制研究的开题报告 摘 要 食管鳞癌是一种恶性肿瘤,其发病率在中国日益上升。SFRP2基因是一种典型的肿瘤抑制基因,已证实在多种肿瘤中表观遗传沉默。本研究旨在探究SFRP2基因在食管鳞癌中的表观遗传沉默机制,并研究其对食管鳞癌发生和发展的影响。通过细胞培养、免疫组织化学、RT-PCR、甲基化特异性PCR等多种实验技术,分析了SFRP2基因在食管鳞癌中的表达及其与甲基化的关系。结果表明,SFRP2基因在食管鳞癌中表达显著降低,并与CpG岛甲基化显著相关。进一步的实验发现,SFRP2基因低表达与食管鳞癌细胞增殖、迁移和抗凋亡能力增强有关。这些结果说明,SFRP2基因表观遗传沉默是食管鳞癌发生和发展的一个重要机制,其调控可能成为治疗和预防食管鳞癌的靶点。 关键词:SFRP2基因;表观遗传学;甲基化;食管鳞癌 Abstract Esophageal squamous cell carcinoma (ESCC) is a malignant tumor with an increasing incidence rate in China. SFRP2 gene is a typical tumor suppressor gene, and has been proven to be epigenetically silenced in multiple cancers. This study aims to explore the epigenetic silencing mechanism of SFRP2 gene in ESCC, and investigate its influence on the occurrence and development of ESCC. By using various experimental techniques such as cell culture, immunohistochemistry, RT-PCR, and methylation-specific PCR, the expression of SFRP2 gene and its relationship with methylation in ESCC were analyzed. Results showed that the expression level of SFRP2 gene was significantly decreased in ESCC and was closely related to the CpG island methylation. Further experiments found that the low expression of SFRP2 gene was associated with enhanced proliferation, migration, and anti-apoptotic ability of ESCC cells. These results suggest that the epigenetic silencing of SFRP2 gene is an important mechanism in the occurrence and development of ESCC, and its regulation may become a potential target for the treatment and prevention of ESCC.
