壳聚糖温敏凝胶对人牙龈成纤维细胞的生物学作用

【摘要】目的:了解壳聚糖温敏凝胶对人牙龈成纤维细胞(GFCs)的生物学作用, 为其局部应用治疗牙周病提供理论依据。方法:人牙龈成纤维细胞在不同浓度的壳聚糖温敏凝胶中体外培养,采用MTT法检测其对GFCs的增殖状况的影响;碱性磷酸酶(ALP)检测试剂盒检测细胞ALP活性以反映壳聚糖温敏凝胶对GFCs分化状况的影响。结果: 各浓度壳聚糖温敏凝胶浸提液对人牙龈成纤维细胞生长和增殖的促进作用明显强于阴性对照组( P 0 . 05)。结论: 壳聚糖温敏凝胶对人GFCs有一定的生物学效应,用于牙周病局部治疗,具有进一步的研究价值。

Biological Effects of the Chitosan Thermosensitive Hydrogel on Human Gingival Fibroblasts Cells in Vitro.

【Abstract】Objective:To evaluate the the biological effects of the chitosan thermosensitive hydrogel on human gingival fibroblasts (HGFs) and provide information about local application of the chitosan thermosensitive hydrogel on periodontal diseases. Methods:Human gingival fibroblasts were cultured in medium containing the chitosan thermosensitive hydrogel at different concentrations,MTT colorimetric assay were used to evaluate the proliferation and ALP kit was used to evaluate the ALP activity of the GFCs. Results: The promotion of different concentrations of the chitosan thermosensitive hydrogel on GFCs growth and proliferation was better than those of control group,through the statistic analysis,there is statistical difference between the 4 groups and the negative control group(P0.05).Conclusion:The results suggest that the chitosan thermosensitive hydrogel could promote the proliferation of the GFCs, local application of the chitosan thermosensitive hydrogel for treating periodontal diseases is feasible.

壳聚糖温敏凝胶系统是一种使用方便、有望在多个领域应用的优异的载药系统,近年来受到高度关注[1-4]。其主要成分是天然高分子材料壳聚糖,室温条件下该系统呈生理中性的溶液,当温度升高到37℃时,该系统可以迅速发生相转变,成为半固态的水凝胶。该载体材料已经被研究用于关节修复、肿瘤治疗、长效镇痛等,但能否用于牙周病治疗以及如何用于牙周病治疗都属于研究空白。
牙龈成纤维细胞( gingival fibroblast cells, GFCs)来源广泛,容易获得,是牙周组织工程的重要种子细胞[ 5 ] 。本研究自制壳聚糖温敏凝胶,并观察其对体外培养的人GFCs的生物学作用,从而为其在临床上的开发应用提供一定的理论
