Title:ConstructionofPetroleumMicrobialGeneticallyEngineeredStrains

Introduction:

Petroleumisanessentialenergysourceandavitalrawmaterialforvariousindustries.Microbialdegradationofpetroleumhasbeenwidelystudiedduetoitspotentialbioremediationapplications.Microorganismssuchasbacteria,fungi,andalgaeplayasignificantroleindegradingthehydrocarbonspresentinpetroleum.However,thenaturaldegradationprocessmaynotbeefficientenoughtomeettheincreasingdemandforpetroleum-basedproducts.Therefore,geneticengineeringisapromisingapproachtoimprovethemicrobialdegradationofpetroleum.

Objectives:

Themainaimofthisprojectistoconstructgeneticallyengineeredstrainsofpetroleum-degradingmicroorganisms.Thespecificobjectivesareasfollows:

1.Identificationandisolationofpetroleum-degradingmicroorganisms

2.Cloningofgenesrelatedtopetroleumdegradation

3.Constructionofexpressionvectors

4.Transformationofthevectorsintotheselectedmicroorganisms

Methodology:

1.IdentificationandIsolationofpetroleum-degradingmicroorganisms

Petroleum-contaminatedsoilsampleswillbecollectedandculturedonnutrientagarmedia.Thegrowthofmicroorganismswillbemonitored,andtheselectedcolonieswillbeidentifiedby16SrRNAsequencing.

2.Cloningofgenesrelatedtopetroleumdegradation

Genesinvolvedinthemetabolismofdifferenthydrocarbonswillbeidentifiedfromtheliterature.PCRprimerswillbedesignedforamplificationofgenesfromgenomicDNAofmicroorganisms.Theamplifiedgenefragmentswillbeclonedintoappropriatevectorsforfurtheranalysis.

3.Constructionofexpressionvectors

Theidentifiedgeneswillbeinsertedintoanexpressionvectorunderthecontrolofapromotertoensureadequateexpression.Transcriptionalandtranslationalterminatorswillbeaddedtotheconstructtoensuregeneexpressionisstoppedatspecificendpoints.
