影响糖尿病小鼠模型血糖的因素考察

[摘 要] 目的:糖尿病模型。方法:ALX的剂量、小鼠的体重、垫料、是否禁食以及禁食时间对小鼠FBG(空腹血糖)的影响,将FBG≥11.1 mmol/L的小鼠模型作为建模成功,计算各组小鼠的成模率和死亡率。结果:注射ALXmg/kg,夜间禁食12~14 h,无垫料的情况下,可成功构建糖尿病小鼠模型。结论:成高,死亡率低,稳定。nfluential factors of blood glucose of?diabetic mouse models

[ABSTRACT] Objective:To investigate the influential factors of blood glucose on diabetic mice models induced by alloxan. Method: A total of 120 Kunming mice were randomly divided into control group and five model groups. Mice in the control group was peritoneally injected with saline at the dose of 10 ml/kg. Meanwhile, mice in the 5 model groups were peritoneally injected with different doses(250,200,150,100,50 mg/kg) of alloxan, respectively. The influence of doses of alloxan, weight of mice, cushioning materials, breeding environment,fasting and fasting time on fasting blood glucose(FBG) were determined. The mice with FBG≥11.1 mmol/L were established as diabetic models. Results: Diabetic mice models can be successfully established using 22~28 g mice, injecting 100~150 mg/kg alloxan, and then fasting 12~14 h in the night and using no cushioning materials. Conclusion: In such conditions, diabetic mice models can be obtained with high success rate, low mortality incidence and stable blood glucose.

糖尿病由多种病因引起的以慢性高血糖为特征的代谢紊乱伴有因胰岛素分泌和作用缺陷引起的糖、脂肪和蛋白质代谢异常。lloxan,ALX)诱导法来制备糖尿病,考察了诱导过程中各因素对模型制备的影响,从而确定了最佳方案。
1 材料和方法
1.1 实验材料
1.1.1 实验动物 180只雄性昆明种小鼠实验动物中心提供动物证1.1.2 仪器 乐康全血糖测定仪(罗氏诊断公司产品,型号2005-06;电子分析天平(梅特勒—托利多仪器(上海)有限公司,型号:AL204)。
1.1.3 药品 ALX由美国Sigma公司腹腔注射ALX 250、200、150、100、50 mg/kg。注射后48 h内灌胃给予10%葡萄糖水,以防止小鼠因低血糖死亡[2]。72 h后禁食12 h,尾取血测血糖,11.1 mmol/L的,小鼠腹腔注射ALX mg/kg,造模后48 h内灌胃给予10%葡萄糖水,3 d后,,测量FBG的变化规律 对1.2.4中的糖尿病小鼠,每周测量一次血糖,连续5周。观察小鼠血糖变化情况,计算平均值。
1.3 统计学处理 采用SPSS 18.
