 SiRNA 干扰 Prohibitin 基因表达对人前列腺 癌 PC-3 细胞生长及凋亡影响# 1,2 1 1 1 5 10 15 20 25 30  (1. 华南肿瘤学国家重点实验室,中山大学肿瘤防治中心泌尿外科, 广东 广州 510060; 2. 广东省泌尿外科重点实验室 (2010A060801016),广东 广州 510060) 摘要: 目的:观察 RNAi 抑制前列腺癌 PC-3 细胞中的 PHB 表达对细胞增殖、凋亡影响,探 讨 PHB 对人前列腺癌的可能作用机制。方法:SiRNA 基因沉默 PC-3 细胞中 PHB 基因,Annexin V/PI 双染和流式细胞计数观察 NM 和 CDM 培养下及分别给予血小板源性生长因子 PDGF 或 EGF 刺激的前列腺癌凋亡细胞数目差别 。结果:流式细胞仪(FCM)检测发现,在 siRNA 抑制 PC-3 细胞中的 PHB 后,CDM 实验组细胞凋亡率显著增加,同 NM 对照组比较有显著性差异(p 0.05 );在 CDM 组给予 PDGF 或 EGF 后,细胞凋亡率升高,与 NM 对照组相比较有显著性差 异(p 0.05 )。结论:低胆固醇培养条件下前列腺癌细胞生长抑制,给予生长刺激因子促进 细胞凋亡,低胆固醇抑制前列腺癌 PC-3 细胞体外增殖活性可能同 PHB 表达升高有关。 关键词:Prohibitin,RNAi,前列腺癌细胞,胆固醇,细胞凋亡 中图分类号:请查阅《中国图书馆分类法》 Effects of SiRNA prohibitin expression on proliferation and apoptosis in human prostate carcinoma PC-3 cells. dongpei1,2, jianglijuan1, guoshengjie1, zhoufangjian1 (1. Guangdong Key Laboratory of Urology,The first affiliated Hospital of Guangzhou medical University, Guangzhou, Guangdong, 510060) Abstract: Objective:To observe the effects of RNAi inhibit PHB on cell growth,and apoptosis in Pc-3 cells.Explore the possible molecular mechanism of PHB on human prostate cancer.Methods:SiRNA knockdown PHB in PC-3 cells and treated cells with NM or CDM added/or not PDGF /EGF. Apoptosis was determined by Annexin V/PI double staining technique and flow cytometry.Results: Annexin V/PI double staining and flow cytometry discovered that after using siRNA knockdowned PHB in PC-3 cells,PC-3 cells apoptosis rate of cultured in CDM was significantly higher than cells cultured in NM(p0.05).Addred PDGF or EGF into CDM can upon apoptosis rate(p0.05 ).Conclusions:Cholesterol depleted conditions can induce apoptosis in PC-3 cells.
