HPLC同时测定藏药圆柏和刺柏中3种黄酮的含量 武雪1,宋平顺2,赵建邦3 (1.兰州大学药学院2012级硕士研究生,兰州730000;2. 3.甘肃省药品检验研究院,甘肃省中药品质与安全评价工程技术研究中心,兰州730070) [摘要] 目的:建立了HPLC法测定藏药圆柏和刺柏中3种主要黄酮成分的方法。方法:采用HPLC,样品经70%甲醇超声,采用色谱柱C18(4.6mm×250mm,5μm),0.2%磷酸乙腈-0.2%磷酸水为流动相进行液相色谱梯度洗脱,流速1.0ml·min-1,检测波长360nm,柱温:40℃。结果:进样量芦丁在0.0049~0.1464μg(R2 =0.9998);槲皮苷在0.0094~0.2814μg(R2 =0.9997);山奈酚在0.0054~0.1620μg(R2 =0.9996)均成良好的线性关系;芦丁、槲皮苷和山奈酚的加样回收率分别为100.12%(RSD2.55%),96.35%(RSD0.72%),96.13%(RSD0.49%)(n=6)。结论:3种黄酮在40min内达到基线分离。该方法分析时间短、稳定性和准确度良好,对圆柏和刺柏药材的质量控制具有一定的参考价值。 [关键词] 藏药; 圆柏;刺柏;高效液相色谱;黄酮;芦丁;槲皮苷;山奈酚 Determination of Three Main flavonoids in??Sabina chinensis and Juniperus formosana by HPLC Wu Xue1 ,Song Ping-Shun2,Zhao Jian-Bang3,(1.2012 Master in Reading,College of Pharmacy in Lanzhou University,Lanzhou73000;2,3.Gansu Institute for Food and Drug Control,Gansu Traditional Chinese Medicine Quality and Safety Evaluation Engineering Research Center, Lanzhou 73000) [Abstract] Objective:A HPLC method has been established for determination of main flavonoids in Sabina chinensis and Junipeius formosana. Method:Samples were extracted in 70% methanol with ultrasonic process. The HPLC was performed on an ODS-C18 column(4.6mm×150mm,5μm)with 0.2% H3PO4acetonitrile-0.2% H3PO4 as mobile phase. The flow rate was 1.0ml·min-1and the column temperature was kept at 40℃. The detective wavelength was 360 nm. Result:The linear range of rutin was 0.0049~0.1464μg(R2=0.9998); the linear rang of quercetin-3-rhamnoside was 0.0094~0.2814μg(R2=0.9997)and the linear range of kaempferol was 0.0054~0.1620μg(R2=0.9996). The average recovery of rutin; quercetin-3-rhamnoside and kaempferol were 100.%(RSD%),%(RSD%),(n=6Conclusion:The results showed that three Flavonoids reached baseline separation within 40min. This method has advantages of using little analytic time, a good stability and accuracy of operation, has some references value for establishing standardization quality control method of Sabina chinensis and Junipeius formosana herb.
