丹参川芎嗪注射液对妊娠期高血压模型大鼠的疗效及胎盘细胞Cyclin B1和p21蛋白的影响 摘要:目的 探讨丹参川芎嗪注射液对妊娠期高血压大鼠模型的效果以及对胎盘细胞中CyclinB1、p21蛋白表达量的影响。方法将孕鼠随机分为4组即:正常妊娠大鼠作空白对照、妊娠高血压大鼠作阴性对照、硫酸镁治疗组妊娠高血压大鼠作阳性对照、丹参川芎嗪注射液组为实验组,每组16只。妊娠14天时,尾静脉注射亚硝基左旋精氛酸甲酯建立妊娠期高血压模型。妊娠第14天开始给药,空白对照组及阴性对照组均给予与药物同剂量的生理盐水;阳性对照组及实验组给予硫酸镁及丹参川芎嗪注射液治疗,3天为一个给药疗程。通过大鼠套尾法测量血压,尾静脉取血法测定24h尿蛋白,并观察各组妊娠大鼠所新生鼠的身长、体重、死胎率、后肢畸形率和胎盘重量,并通过免疫蛋白印迹法测定各组大鼠胎盘细胞中CyclinB1、p21蛋白表达量。结果 建立模型之后,孕鼠尿蛋白以及血压增加,新生鼠的身长、体重和胎盘重量均显著降低,死胎率以及畸形率显著增高;给药治疗后,阳性对照组以及实验组四组孕鼠胎盘组织细胞cyclinB1以及p21浓度,与空白对照组相比较,其他三组患者cyclinB1蛋白浓度均减小(P0.05),p21浓度显著增高 (P0.05),阳性对照组与实验组相比较,差异无统计学意义(P0.05)。尿蛋白以及血压减低,新生鼠的身长、体重和胎盘重量均显著升高,差异具有统计学差异(P0.05),阳性对照组与实验组相比较死胎率以及畸形率具有统计学差异(P0.05)。结论 川芎嗪注射液能够显著降低妊娠高血压孕鼠的死胎率以及畸形率,并且新生鼠状况也有所好转,并且能够激活cyclinB1以及下调p21浓度,这可能是川芎嗪注射液治疗效果的原因之一。 关键词:丹参川芎嗪注射液;妊娠期高血压;CyclinB1蛋白;p21蛋白 [中图分类号] TQ460. 7??
